3 Method summary
The sample remaining in the clavulanic acid with pH6.0 Tris (Tris) buffer - acetonitrile extraction, dichloromethane - n-hexane
Liquid-liquid partition purification, molecular sieve ultrafiltration, liquid chromatography - mass spectrometry/mass spectrometry detection and confirmation, external standard.
4 Reagents and materials
Unless otherwise specified, the reagents used were of analytical grade water as a water line with GB/T 6682 regulations.
4.1 Methanol. HPLC grade.
4.2 acetonitrile. HPLC grade.
4.3 dichloromethane.
4.4 n-hexane.
4.5 acidic alumina.
4.6 Tris (Tris).
4.7 hydrochloric acid.
5mmol/LTris of 4.8 pH6.0 buffer solution. Weigh 0.61gTris (4.6), was dissolved with about 900mL of water (4.7) adjusted with hydrochloric acid
pH value 6.0, set the volume to 1000mL
4.9 clavulanate potassium (clavulanicacidpotassium, CAS number. 61177-45-5) standards, the purity of not less than 99%.
4.10 standard stock solution, 5000mg/L. Weigh 59.5mg potassium clavulanate (equivalent to 50mg clavulanic acid) standard (4.9),
(4.8) dissolved 5mmol/LTris pH6.0 buffer solution of a small amount, and then acetonitrile (4.2) and dilute to 10mL volumetric flask, at -30 ℃
