3 Method summary

4 Reagents and materials
Unless otherwise indicated, the reagents used were of analytical grade water as a water GB/T 6682 regulations.
4.1 acetonitrile. HPLC grade.
4.2 Methanol. HPLC grade.
4.3 TCA.
4.4 of concentrated aqueous ammonia (25%).
4.5 ammonium acetate.
4.6 5% TCA. TCA accurately weighed 50g (4.3), add water, 950mL dissolve, mix well.
Methanol + water 4.7. 1 + 1 (volume ratio).
Methanol + aqueous ammonia 4.8. 4 + 1 (volume ratio).
4.9 pH9.250mmol/L ammonium acetate solution. Accurately weigh 3.854g of ammonium acetate (4.5), add 900mL water dissolved, aqueous ammonia (4.4)
Adjusting the pH to 9.2, transferred to 1000mL volumetric flask and water to volume, and mix well.
4.10 sample volume solution. take 95mL50mmol/L ammonium acetate solution (pH9.2) (4.9) was mixed with 5mL methanol uniform.
