1 Scope
This standard specifies the sample preparation and liquid chromatography ammonia nitrogen in foods of animal origin phenanthridine residues - MS/MS detection method.
This standard applies to beef, beef liver, beef kidney, beef fat, goat, chicken, fish and milk ammonia nitrogen phenanthridine residues measured and confirmed.
2 Method summary
Ammonia nitrogen in the sample phenanthridine with acetonitrile and methanol mixed solution of ammonium formate extract, the extract was concentrated and defatted by liquid chromatography - mass spectrometry /
Mass spectrometer, external standard peak area quantification.
3 Reagents and materials
All reagents unless otherwise specified, were of analytical grade, double distilled water.
3.1 Methanol. HPLC fractions.
3.2 formic acid. HPLC fractions.
3.3 acetonitrile. HPLC fractions.
3.4 n-hexane.
3.5 anhydrous sodium. 650 ℃ burning 4h, cooled in a desiccator to room temperature and stored in a sealed bottle aside.
3.6 Ammonium. excellent pure or equivalent person.
3.7 Ammonium formate - methanol solution (0.25mol/L). Weigh 15.8g of ammonium formate was dissolved in 1000mL methanol (3.1) in.
3.8 80% methanol solution. Methanol (3.1) 800mL, diluted with water to 1000mL, and mix.
Extract 3.9. Take acetonitrile (3.3) and ammonium formate - methanol solution (3.7) were mixed 1 (volume ratio).
