1 Scope
This standard specifies the method of enzyme-linked immunosorbent assay of total import and export food residues in α- and β- endosulfan endosulfan.
This standard applies to cereal grains, vegetables, tea, tobacco, fruits, meat and meat products, shrimp endosulfan residues screening tests. Fish NA

2 Determination
2.1 Method summary
Extracting samples α- and β- endosulfan endosulfan with methanol or acetone, detected by competitive ELISA.
2.2 Reagents and materials
Unless otherwise requested, all reagents were of analytical grade, double distilled water.
2.2.1 methanol.
2.2.2 acetone.
2.2.3 Potassium ferrocyanide 0.36mol/L.
2.2.4 zinc sulfate solution 1.04mol/L.
2.2.5 disodium hydrogen phosphate (Na2HPO4 · 12H2O).
2.2.6 sodium dihydrogen phosphate (NaH2PO4 · H2O).
2.2.7 sodium chloride.
2.2.8 anhydrous sodium acetate.
2.2.9 β- cyclodextrin.
2.2.10 urea hydrogen peroxide.
2.2.11 3,3 ', 5,5'-tetramethyl benzidine.
2.2.12 dimethyl sulfoxide.
2.2.13 phosphate buffer (pH7.4). See Appendix A.
2.2.14 Tween-20.
2.2.15 anhydrous sodium. 650 ℃ burning 4h, cooled in a desiccator to room temperature and stored in a sealed bottle aside.
2.2.16 0.5% fish skin glue solution (stored at 4 ℃).
2.2.17 masking agents dilution. 0.5% gum skin. phosphate buffer (1.20, by volume), and diluting at the time of use.
2.2.18 Solution I. methanol. masking agent dilutions (1.10, by volume).
2.2.19 enzyme markers. hapten and horseradish peroxidase conjugate, 4 ℃ preservation, with phosphate buffer in the use of 40-fold dilution,

2.2.21 substrate material solution A, see Appendix A.
2.2.22 substrate solution B, see Appendix A.
2.2.23 Stop Solution. sulfuric acid 1.25mol/L.
2.2.24 α, β- Dan mixing sulfur standards (English name. α, β-Endosulfan, CAS Code. 115-29-7). not less than 99.5% purity.
2.2.25 Standard stock solution. formulated as 1μg/mL stock solution in methanol, -20 ℃ preservation.
