SN/T 1692 is divided into three parts.

1 Scope
This part of SN/T 1692 specifies the procedure for the African Stirrup Complement Binding Test.
This section applies to the inspection and quarantine of African horse stables.

GB/T 6682 Analytical laboratory water specifications and test methods
3 reagents and materials
The water used in this section should meet the specifications of the third grade water in GB/T 6682. The reagents used are analytically pure reagents unless otherwise specified.
3.1 African Stirrup Anti-Antigen. Provided by the designated unit or prepared according to Appendix A.
3.2 Complement. Provided by the designated unit or prepared in accordance with Appendix A.
3.3 Hemolysin. provided by the designated unit.
3.4 Positive serum. provided by the designated unit, diluted 1.2 before use, and extinguished in a water bath at 58 °C ~ 59 °C for 30 min.
3.5 Negative serum. provided by the designated unit, diluted 1.10 before use, and extinguished at 58 ° C ~ 59 ° C water bath for 30 min.
3.6 Alzheimer's solution. Prepared according to Appendix A.
3.7 Barbiturate Buffer (VBS). Prepared according to Appendix A.
4 instruments and equipment

5 sample processing
After the animal number to be inspected is registered, the blood is collected aseptically, centrifuged at 5000 r/min 10 ° C for 10 min to 15 min, and the serum is separated aseptically.
Store at -70 ° C (for a formal test, one spare), use 1.100 dilution with barbital buffer before use, horse serum 56 ° C, zebra blood
Clear 60 ° C, sputum serum 62 ° C water bath for 30 min inactivated.
6 preliminary test
6.1 2% sheep red blood cell suspension preparation
6.1.1 Take one sterilized flask containing 100 mL of albino solution, and collect 50 mL of sterile blood from the jugular vein of the ram. After mixing, set the refrigerator at 4 °C.
Set 4d ~ 5d.
6.1.2 When inhalation, inhale the blood into the centrifuge tube, centrifuge at 1500r/min for 15min, and use a pipette to remove the supernatant and white blood cells from the upper layer of red blood cells.
The platelet membrane was washed with BBS buffer, centrifuged at 1500 r/min for 15 min, and the supernatant was discarded, and the washing was repeated four times.
6.1.3 Dilute the last washed red blood cell sludge with VBS buffer at 2% (2mL cells + 98mL VBS)
