3 reagents and materials

3.1 Antigen. African horse stable tissue culture antigen. 3.2 African horse stable positive serum. 3.3 plate. 3.4 Micropipettes. 3.5 micro dripper. 3.6 Erlenmeyer flask. 3.7 agarose. 3.8 Sodium chloride. analytically pure. 3.9 Sodium azide. analytically pure. 3.10 Water. Meet the secondary water specifications of the GB/T 6682 analytical laboratory.

4 steps

4.1 Preparation of agar gel plate Weigh 1.2g of agarose, 1.0g of sodium chloride, add 100mL of water, dissolve it by high pressure or boiling after 68.94kPa, 10min, then cool to At 45 ° C, 0.01 g of sodium azide was added. Add agar to each plate and make the agar plate thickness 2.5mm~2.8mm, and then invert after solidification. Put in a refrigerator at 4 ° C for 15 min. 4.2 punching 1 hole in the middle and 6 holes in the surrounding group. The aperture is 4 mm and the hole spacing is 2.4 mm. Use a needle to pick out or aspirate the agar in the well. 4.3 Loading The central well was added with antigen, 1, 3, and 5 wells plus standard positive serum; 2, 4, and 6 wells were added to the test serum. The amount of sample is based on the hole level, and the sample is placed after the wet box. The reaction was carried out at room temperature (22 ° C to 25 ° C), and a positive control was set for each test.

5 judgment

5.1 24h initial judgment, 72h final judgment. When the positive control is established, a determination is made. Positive. a dense sediment line appears between the test serum well and the antigen well, and is smooth with the end of the sedimentation line of the standard positive serum. Connected.
