Determination of polyphenols in vegetable oils by grain and oil
1 Scope
This standard specifies the principle, reagents, instruments, sample preparation, operation steps and knots for the determination of polyphenol content in vegetable oil by spectrophotometry.

This standard applies to the determination of polyphenol content in vegetable oils.
The detection limit of this standard is 6mg/kg.

3 Principle
After the polyphenols in the vegetable oil are purified by the diol-based cartridge, the forin phenol reagent oxidizes the -OH group in the polyphenol and is blue, at the maximum absorption wavelength.
The absorbance of the solution was measured at 750 nm, and the polyphenol content in the vegetable oil was quantitatively determined using gallic acid as a calibration standard.

Analytically pure reagents should be used in the analysis unless otherwise stated. Water is the recommended tertiary water for GB/T 6682.
4.1 Methanol. Chromatographically pure.
4.2 Hexane. Chromatographically pure.
4.3 Methanol-water solution. Accurately transfer 50 mL of methanol (4.1) to a 100 mL volumetric flask and bring up to volume with water.
4.4 Sodium carbonate.
4.5 Folin-ciocalteu reagent.
4.6 7.5% sodium carbonate solution (mass concentration). Weigh 7.5g ± 0.01g sodium carbonate (4.4), add appropriate amount of water to dissolve, transfer to 100mL capacity

4.7 Gallic acid standard. purity ≥ 98%, relative molecular mass 170.12.
4.8 Gallic acid standard stock solution (1mg/mL). Weigh 0.100g ± 0.001g gallic acid standard (4.7), add a small amount of methanol
