Inhibition zone method1 ScopeThis standard specifies the method for determining the zone of inhibition for the antibacterial properties of peptides.
This standard applies to the determination of antibacterial properties and anti-filamentous fungi properties of polypeptides.2 Normative referencesThe following documents are indispensable for the application of this document. For dated reference documents, only the dated version applies to this article
Pieces. For undated references, the latest version (including all amendments) applies to this document.
GB 4789.2 National Food Safety Standard, Food Microbiological Examination, Determination of Total Colony
GB/T 6682 Analytical laboratory water specifications and test methods3 Terms and definitionsThe following terms and definitions apply to this document.
3.1
Polypeptides
Between amino acids and proteins, a class of compounds formed by two or more amino acids connected by peptide bonds.
3.2
Inhibition zone
The zone of aseptic propagation at the boundary between the surface of the solid medium and the sample.
3.3
Antimicrobial activity
The ability to inhibit the reproduction of microorganisms is expressed by the antibacterial titer U value (AU).4 PrincipleAfter the polypeptide is in contact with the test indicator bacteria in the solid plate, the growth of the surrounding strains is inhibited, forming a sterile breeding area,
According to the diameter of the inhibition zone of the test bacteria on the surface of the solid plate, calculate the antibacterial titer U value to determine the activity.5 Reagents or materialsUnless otherwise specified, the reagents used in this method are analytically pure, and the experimental water is the first grade water specified in GB/T 6682.
5.1 Normal saline
0.85% normal saline. Weigh 8.5g of sodium chloride in a volumetric flask, dissolve it with first-grade water and dilute to 1000mL. 121℃±1℃
Autoclave sterilization for 20 minutes.
