The main difference between this and the standard EPA methods are.

--- Incubator temperature from 35 ℃ ± 0.5 ℃ revised to 36 ℃ ± 1 ℃;

1 Scope
This standard specifies the method for determining entry and exit procedures and the ship's ballast water and coliform bacteria (coliforms) using membrane filtration method.
This standard applies to the entry and exit of ships' ballast water and coliform counts.
2 Equipment and Materials
2.1 incubator. 36 ℃ ± 1 ℃.
2.2 Refrigerator. 2 ℃ ~ 8 ℃.
2.3 100mL graduated cylinder. sterilization.
2.4 membrane filtering device 1). see 4.1.2.

2.5 Sterile cellulose ester membrane, white, with a grille, 47mm diameter, pore size 0.45μm.
2.6 Sterile pipettes. 10mL, accurate to 0.1mL.
2.7 sterile saline.
2.8 conical flask. 150mL.
2.9 UV lamp. wavelength 366nm.
2.10 sterile Petri dish. a diameter of 50mm.
2.11 sterile filter. pore size of 0.22μm.
2.12 MI medium. See Appendix A.

4.1 Preparation
4.1.1 Preparation of MI agar medium, preservation medium should be 4 ℃ temperature returned to room temperature before use.
