1 Scope
This standard specifies the mosquitoes insensitive acetylcholinesterase (AChE) detection method.
This standard applies to Culex pipiens (Culexpipienspallens), Culex quinquefasciatus (Cx.pipiensquinquefasciatus), Aedes
Mosquito (Aedesalbopictus), Anopheles (Anophelessinensis) and other organophosphate, carbamate insecticides resistant organisms

2 Terms and definitions
The following terms and definitions apply to this document.
2.1
Acetylcholinesterase distinction concentration discriminatingconcentrationofacetylcholinesterase
It can inhibit the mosquito acetylcholinesterase sensitive physiologically active concentration of 90% insecticide.
3 test insects

4 Reagents
4.1 disodium hydrogen phosphate (Na2HPO4), AR.
4.2 potassium dihydrogen phosphate (KH2PO4), AR.
4.3 pull-through X-100 (TritonX-100).
4.4 acetylthiocholine (ATCh), AR.
4.5 5,5'-dithiobis-nitrobenzoic acid (DTNB), AR.
4.6 phosphate buffer (0.1mol/L, pH7.5PBS). Weigh 30.18gNa2HPO4 · 12H2O and 2.16gKH2PO4, distillation
Water volume to 1000mL.
4.7 DTNB- reagent ethanol. 12.4mgDTNB, plus 120mL95% ethanol, 80mL of distilled water, 50mL phosphate buffer
(PH7.5,0.1mol/L, containing 0.1% TritonX-100).
4.8 ATCh solution. Weigh 28.9mgATCh dissolved in 10mL distilled water, -20 ℃ stored.
4.9 ATCh-DTNB mixed solution. Weigh 75mgATCh and 13mgDTNB, dissolved in 100mL phosphate buffer.
