3.1 Model Nomenclature

4 Requirements
4.1 Slides and coverslips, appearance, specimen collection and processing, specimen staining, and specimen covers should comply with JY/T 0067-2011
Chapter 4.
4.2 Specimens should be taken from the lower epidermis of fresh broad-stemmed broad bean leaves.
4.3 Specimens should be tiled pieces, each piece of material should be not less than 2mm × 2mm, cut around neatly.
4.4 It should be possible to see the irregular epidermal cells and their nuclei and the stomata scattered between the epidermal cells.
4.5 The morphology of the guard cells should be normal, and the nucleus and chloroplast should be clearly visible.
4.6 The materials are neat and the attached mesophyll or dirt shall not affect the observation.
4.7 The closed air hole shall not exceed 2/3.
4.8 The cytoplasm is uniformly colored, the nuclei are obvious, and the cell boundaries are clear.
5 Test method
5.1 According to the provisions of Chapter 5 of JY/T 0067-2011.
5.2 Specimens are examined at a microscope magnification of 100 times.
