This standard was drafted in accordance with the rules given in GB/T 1.1-2009 "Guidelines for Standardization Work_Part 1. Structure and Preparation of Standards"
This standard replaces JY 70-1982 "Technical Conditions for Top Bud Slitting". In addition to editorial changes, the main technical changes are as follows.
-Eliminate the "slice thickness within 8μm" which is difficult to inspect;
-Elimination of single dyeing requirements;

1 Scope
This standard specifies the type designation, requirements, test methods, inspection rules, marks, instructions, packaging, and transportation of the top bud longitudinal section.

4 Requirements
4.1 Slides and coverslips, appearance, specimen collection and processing, specimen staining, and specimen covers should comply with JY/T 0067-2011

4.2 Specimens should be taken from the top buds of Hydrilla verticillata, and the middle part of the buds should be cut longitudinally, and one piece of material is placed on each slide.
4.3 The growth cone, leaf primordia, young leaves, axillary bud primordia, and bud axis are clearly shown.
4.4 The original set of cells in the outermost layer of the growth cone should be neatly arranged. The original cells in the original set are not aligned and have a large volume.
4.5 There should be no obvious "plasma separation" in the cells of the growth cone and young leaves.
4.6 The young leaves should be completely wrapped on the growth cone.
