1 Scope
This standard specifies the sample preparation and liquid chromatography imported and exported food dinotefuran residues - MS/MS detection method.
This standard applies to wheat, peanuts, corn, spinach, apples, carrots, basil, pork and salmon in dinotefuran residues detected
And confirmation.
2 Method summary
Samples were extracted with acetonitrile, after the extract was added over anhydrous sodium sulfate, graphite on non-porous carbon column (Envi-Carb)/amide propyl silyl
Alkylated silica gel column (LC-NH2) purification, liquid chromatography - mass spectrometry/mass spectrometry, external standard.
3 Reagents and materials
Unless otherwise specified, reagents were HPLC grade water for ultra-pure water.
3.1 acetonitrile.
3.2 n-hexane.
3.3 acetone.
3.4 ether.
3.5 Sodium chloride. pure class distinctions.
3.6 anhydrous sodium sulfate. AR, 650 ℃ burning 4h, natural cooling spare stored in a sealed bottle.
3.7 dipotassium hydrogen phosphate (K2HPO4). AR.
3.8 potassium dihydrogen phosphate (KH2PO4). AR.
3.9 Sodium hydroxide. AR.
3.10 hydrochloride. pure class distinctions.
3.11 1mol/L sodium hydroxide. Weigh 40g of sodium hydroxide (3.9), was dissolved in 1L of water.
3.12 1mol/L hydrochloric acid. Weigh 36.5g of pure hydrochloric acid (3.10), was dissolved in 1L of water.
3.13 hexane saturated acetonitrile. in 250mL separating funnel were added 100mL of acetonitrile (3.1) and 100mL hexane
(3.2), fully shaking, standing layer, the lower layer of n-hexane saturated acetonitrile.
3.14 acetone - n-hexane (1 + 1, volume ratio). An equal volume of acetone (3.3) and n-hexane (3.2) were mixed sufficiently shaken.
3.15 phosphate buffer solution. 0.5mol/L (pH = 7.0), weighed 52.7g and 30.2g of dibasic potassium dihydrogen phosphate, is added about
500mL dissolved in water, adjusted pH7.0 with 1mol/L sodium hydroxide or 1mol/L hydrochloric acid, and add water to 1L.
3.16 dinotefuran Standards. dinotefuran, C7H14N4O3, CAS number. 165252-70-0 purity of greater than or equal to 98%.
3.17 dinotefuran standard stock solution. weigh the amount of dinotefuran standard with acetone - n-hexane (1 + 1, volume ratio) formulated as 1.0mg/mL
Standard stock solution; 0 ℃ ~ 4 ℃ preservation. 1 year shelf life.
3.18 dinotefuran standard working solution. if necessary with acetone - n-hexane (1 + 1, volume ratio) stock solution was diluted to a suitable concentration of the standard preparation
Quasi working fluid. 0 ℃ ~ 4 ℃ preservation. Save 1 month.
3.19 Envi-Carb/LC-NH2 solid phase extraction column. 500mg/500mg. SupelcleanENVI-CarbSPE9 (non-porous graphitized
Carbon) with SupelcleanLC-NH2SPE (amidopropyl silylated silica gel) in series.
3.20 Membrane. 0.2μm.
