1 Scope
This standard specifies the foods of animal origin nitrogen risperidone and its metabolite N piperazine alcohol Residues gas chromatography - mass spectrometry.
This standard applies to pork and kidney by nitrogen and determination Risperidone piperazine nitrogen metabolite of alcohol residues and confirmation.
2 Method summary
Samples were extracted with acetonitrile, purified by C18SPE acidic acetonitrile, n-hexane liquid-liquid partition, after tert-butyl methyl ether, alcohol piperazine nitrogen
Then by trimethylsilyl derivatives by gas chromatography - mass spectrometry, internal standard.
3 Reagents and materials
Unless otherwise specified, the reagents were of analytical grade, water is distilled water.
3.1 methanol. liquid chromatography.
3.2 hexane. liquid chromatography.
3.3 acetonitrile. liquid chromatography.
3.4 ethyl acetate. liquid chromatography.
3.5 tert-butyl ether. liquid chromatography.
3.6 sulfuric acid. 98%.
3.7 sodium hydroxide.
3.8 sodium chloride.
3.9 10% sodium chloride solution. Weigh 100g of sodium chloride dissolved in an appropriate amount of water, and the water volume to 1L.
3.10 sulphate solution. 28mL sulfuric acid (3.6) containing 500mL of water was added slowly in a beaker, cooled to room temperature and mix standby.
3.11 0.01mol/L sulfuric acid solution. Pipette 20mL sulfuric acid solution (3.10) to 1L of water containing 250mL beaker, add water to
1L.
3.12 0.05mol/L sulfuric acid solution. Pipette 100mL sulfuric acid solution (3.10) to water containing 250mL 1L measuring cup, add water to
1L.
3.13 acidic acetonitrile solution. join 1mL0.05mol/L sulfuric acid solution (3.12) in 100mL acetonitrile.
3.14 0.5mol/L sodium hydroxide solution. Weigh 20g sodium hydroxide dissolved in an appropriate amount of water, and the water volume to 1L.
3.15 acetonitrile saturated n-hexane solution. Take 100mL n-hexane, 30mL of acetonitrile in a separating funnel, Shakers, and set aside.
3.16 derivatizing agents. bis-trimethylsilyl trifluoroacetamide (BSTFA) - trimethylchlorosilane (TMCS) (99 + 1, volume ratio).
3.17 2,3,4-2 ', 4', 5'-hexachlorobiphenyl (PCB138). purity greater than or equal to 98%.
3.18 Internal standard solution. ethyl acetate preparation 2,3,4-2 ', 4', 5'-hexachlorobiphenyl (PCB138) at a concentration of 0.10μg/mL.
3.19 azapirone standards (Azaperone, CAS Number. 1649-18-9 suppliers. C19H22FN3O). purity greater than or equal to 98%.
