1 Scope
This standard specifies the positive staphylococci imported and exported food coagulation detection methods.
Counting this standard plate count method is suitable for direct use in food coagulase-positive staphylococci, MPN method is suitable for coagulase-positive
Staphylococcus bacteria content is lower and the higher the content of food. Animal feed and other food available for implementation.
2 Equipment and Materials
2.1 sterilization equipment. high-pressure steam autoclave and dry heat sterilization box.
2.2 water bath. 48 ℃ ± 1 ℃.
2.3 Incubator. 36 ℃ ± 1 ℃.
2.4 straw. 1mL and 10mL, sub-scale were 0.01mL and 0.1mL.
2.5 inoculating loop. 3mm diameter.
2.6 Balance. Range 2kg, the sensitivity of 0.1g.
2.7 Sterilization of sample preparation equipment. sampling spoon, scissors, can opener and so on.
2.8 Sample dilution bottle. 250mL, 500mL.
2.9 sterile Petri dish. diameter of 90mm.
2.10 homogenizer.
2.11 sterile tubes.
2.12 alcohol lamp or gas lamps.
2.13 pH meter.
2.14 Staphylococcus aureus (coagulase positive) Standard quality control strains. ATCC27217 qualified or certified strains from other sources.
3 media and reagents
3.1 rabbit plasma fibrinogen (RPF) agar (see Appendix A A.1 chapter).
3.2 Improved GC broth (modifiedGiolittiandCantonibroth, see Appendix A, Section A.2 chapter).
3.3 Water Agar (see Chapter A.3 Appendix A).
3.4 peptone sodium chloride solution (see Appendix A Chapter A.4).
