Determination of algal fatty acids by gas chromatography
1 Scope
This standard specifies the gas chromatographic method for the determination of fatty acids in algae.
This standard applies to the determination of fatty acids in algae. Determination of fatty acids in other algae can be used as reference.

GB/T 6682 Analytical laboratory water specifications and test methods
3 Principle of the method
The fatty acid in the algae sample is saponified with sodium hydroxide in methanol solution and methyl esterified with methanolic hydrochloric acid solution. After extraction with n-hexane, the gas phase is used.
Chromatographic determination.
4 reagents and materials
Unless otherwise stated, the reagents used in this method are of analytical grade and the water is the first grade water specified in GB/T 6682.
4.1 Reagents
4.1.1 Potassium hydroxide (KOH).
4.1.2 Hydrochloric acid (HCl).
4.1.3 Sodium chloride (NaCl).
4.1.4 n-Hexane (C6H14). Chromatographically pure.
4.1.5 Methanol (CH3OH). chromatographically pure.
4.1.6 anhydrous sodium sulfate (Na2SO4). burning at 400 ° C for 4 h, sealed and stored after cooling, valid for 1 month.
4.2 Reagent preparation
4.2.1 Saturated potassium hydroxide methanol solution. Weigh 40g of potassium hydroxide dissolved in 100mL of methanol, stir to dissolve, clarify standby, room temperature

4.2.2 Hydrochloric acid methanol solution (1mol/L). Dissolve 10mL hydrochloric acid in 110mL methanol, mix and store at room temperature for 2 months.
4.2.3 Saturated sodium chloride solution. Weigh 360g of sodium chloride dissolved in 1L of distilled water, stir to dissolve, clarify and reserve, and place at room temperature.
