7 Analysis steps
7.1 Extraction
Weigh 20 g of sample (accurate to 0.01 g) in a 80 mL centrifuge tube, add 40 mL of acetonitrile, homogenize at 15000 r/min homogenate
Extract 1min, add 5g sodium chloride, and then homogenate extraction 1min, the centrifuge tube into the centrifuge, centrifugation at 3000 r/min 5min, take
Clear liquid 20mL (equivalent to 10g sample volume), to be purified.
7.2 Purification
7.2.1 Envi -18 column into the fixed frame, before adding the first 10mL acetonitrile pre-wash column, then the next heart of the bottle, into the 20mL extraction
And the column was washed with 15 mL of acetonitrile and the collected extract and washings were spin concentrated to about 1 mL in a water bath at 40 ° C for use.
7.2.2 To the Envi -Carb column, add about 2 cm of anhydrous sodium sulfate, connect the column to the top of the Sep-Pak ammonia column,
Then put the bottle on the fixed frame. The column was pre-washed with 4 mL of acetonitrile-toluene solution (3 1) before loading, and when the liquid level reached the top of the sodium sulfate,
The sample concentrate (7.2.1) was quickly transferred to a purification column and the vial was washed three times with 2 mL of acetonitrile-toluene solution (3 1)
And the washing liquid was transferred to the column. A 50 mL reservoir was added to the tandem column and the tandem was washed with 25 mL of acetonitrile-toluene solution (3 1)
All effluent was placed in a chicken heart and concentrated in a 40 ° C water bath to about 0.5 mL. Each time 5 mL of n-hexane was added in a 40 ° C water bath
Rotary evaporation, solvent exchange twice, and finally the sample volume is about 1mL, add 40μL internal standard solution, mix, for gas chromatography
- Mass spectrometry.
