1 Scope
This standard specifies the bovine paratuberculosis enzyme-linked immunosorbent assay method.
This standard applies to the detection of bovine paratuberculosis antibodies.

GB/T 6682 analytical laboratory use specifications and test methods (GB/T 6682-1992, neq ISO 3696)
3 test materials
3.1 Reagents
--- Mycobacterium paratuberculosis antigen-coated plate;
--- Positive control serum;
--- Negative control serum;
--- Horseradish peroxidase (the HRPO) labeled secondary anti-antibody;
--- Sample diluent;
--- Washer fluid;
--- The TMB substrate solution;
--- Stop solution.
The kit should be stored at 2 ℃ ~ 7 ℃. Before use to restore to room temperature (20 ℃ ~ 25 ℃), and mix well.
3.2 serum to be tested
By conventional methods and isolated blood serum at 4 ℃ refrigerator spare. For long-term storage, should be placed in -20 ℃ refrigerator.
4 Equipment and Materials
4.1 applies to 96-well microtiter plate in a microplate reader.
4.2 oscillator.
4.3 100mL, 500mL, 1000mL graduated cylinder.
4.4 8 or 12 washer.
4.5 8 or 12 pipette (50μL ~ 300μL).
4.6 Single-channel pipette (0.5μL ~ 10μL; 40μL ~ 200μL; 200μL ~ 1000μL).
4.7 96-well microtiter plate to dilute the sample.
4.8 refrigerator.
5 Procedure
5.1 dilution of serum to be tested
Serum was seized by the selected kit require dilution, after the oscillator mix well and set aside.
