1 Scope

This standard specifies the method for the determination of toxic levels of biogenic amines in aquatic products by high performance liquid chromatography. This standard applies to shrimp, fish, shellfish meat phenylethylamine, putrescine, cadaverine, histamine, octopamine, tyramine and spermidine detection.

2 Determination

2.1 Method summary Biogenic amines in the sample solution with perchloric acid extraction, dansyl chloride derivatization with UV detector with high performance liquid chromatography separation Detection, external standard. 2.2 Reagents and materials Unless otherwise specified, the reagents were of analytical grade, water is deionized water, with water as the mobile phase of ultrapure water. 2.2.1 Methanol. HPLC grade. 2.2.2 acetonitrile. HPLC grade. 2.2.3 acetone. HPLC grade. 2.2.4 Dan chloride. HPLC grade purity ≥99.0%. 2.2.5 sodium bicarbonate. 2.2.6 perchloric acid. ρ = 1.67g/mL. 2.2.7 sodium hydroxide. 2.2.8 concentrated aqueous ammonia. 25% to 28%. 2.2.9 0.4mol/L perchloric acid solution. Measure 48mL of perchloric acid (ρ = 1.67g/mL), diluted with water to 2L. 2.2.10 2mol/L sodium hydroxide solution. Weigh 4.0g of sodium hydroxide dissolved in 50mL water. 2.2.11 saturated sodium bicarbonate solution. 2.2.12 10mg/mL Dan chloride solution. Weigh 0.20g Dan chloride dissolved in 20mL acetone. 2.2.13 Standard. phenylethylamine hydrochloride (2-Phenylethylaminehydrochloride, C6H5CH2CH2NH2 · HCl, CAS. 156-28-5, purity ≥98%), and putrescine hydrochloride (Putrescinedihydrochloride, NH2 (CH2) 4NH2 · 2HCl, CAS. 333-93- 7, purity ≥98%), hydrochloric acid cadaverine (Cadaverinedihydrochloride, NH2 (CH2) 5NH2 · 2HCl, CAS. 1476-39-7, pure Of > 98.0%), histamine dihydrochloride (Histaminedihydrochloride, C5H9N3 · 2HCl, CAS. 56-92-8, purity ≥99.0%), salt Acid octopamine (Octopaminehydrochloride, HOC6H4CH (CH2NH2) OH · HCl, CAS. 770-05-8, purity ≥ 99.0%), tyramine hydrochloride (Tyraminehydrochloride, HOC6H4CH2CH2NH2 · HCl, CAS. 60-19-5, purity ≥ 99.0%), spermidine hydrochloride (Spermidinetrihydrochloride, NH2 (CH2) 3NH (CH2) 4NH2 · 3HCl, CAS. 334-50- 9, purity ≥99.5%). 2.2.14 mixed standard stock solution. Weigh accurately hydrochloride phenylethylamine, putrescine hydrochloride, hydrochloric acid, cadaverine, histamine dihydrochloride, hydrochloric acid octopamine, tyramine and hydrochloric acid Spermidine hydrochloride standard each 0.130g, 0.183g, 0.172g, 0.166g, 0.124g, 0.127g and 0.176g in 100mL flask, And dissolved in water to volume, to give a mixed standard stock solution 1.00mg/mL.
