3 Overview
Epidemic hematopoietic necrosis (Epizotic haematopoieticnecrosis, EHN) is an epidemic hematopoietic necrosis
Epizotichaematopoieticnecrosisvirus (EHNV) causes finfish to be characterized by systemic clinical or subclinical infections
Viral infections (see Appendix A).
4 reagents and materials
4.1 Water. Meet the specifications of the first grade water in GB/T 6682.
4.2 EHNV reference strain. provided by the designated unit.
4.3 Cell line. Blue scorpion fibroblast cell line (Bluegilfry, BF-2), Fathead 鱥 cell line (Fathead Minnow,
FHM), Epithelioma Papulosum Cyprini (EPC) or Large Scale Salmon Embryo Cell Line
(ChinookSalmonEmbryo, CHSE-214).
4.4 4-Hydroxyethylpiperazineethanesulfonic acid (HEPES).
4.5 Rabbit anti-EHNV antibody or murine anti-EHNV antibody. supplied by the designated unit.
4.6 Horseradish peroxidase-labeled anti-rabbit or anti-mouse immunoglobulin (IgG).
4.7 TaqDNA enzyme. Store at -20 °C to avoid drastic changes in temperature.
4.8 dNTPs. containing 10 mmol/L of dCTP, dGTP, dATP and dTTP.
4.9 Primers. The concentration was 40 μmol/L. The sequence is as follows.
