Detection in food import and export of avermectin residues
High performance liquid chromatography - mass spectrometry/mass spectrometry1 ScopeThis standard specifies the foods HPLC avermectin residues - MS/MS detection method.
This standard applies to rice, garlic, spinach, apples, chestnuts, tea, beef, lamb, chicken, fish, red peony root, vinegar and honey Avi
Avermectin residues detected.2 Method summaryAfter the first sample honey and vinegar diluted with water to a C18 solid phase extraction (SPE); other samples were extracted with acetonitrile, the solid phase with neutral alumina
Extraction (SPE), high performance liquid chromatography - mass spectrometry/mass spectrometry, external standard.3 Reagents and materialsUnless otherwise specified, reagents were analytical grade water for ultra-pure water (greater than 18MΩ).
3.1 acetonitrile. high performance liquid chromatography.
3.2 aqueous acetic acid (0.1%). Take 1mL acetic acid to water volume to 1000mL.
3.3 aqueous acetonitrile. acetonitrile + water (6 + 1, volume ratio).
3.4 anhydrous sodium. 650 ℃ before use burning 4h, cooled in a desiccator to room temperature and stored in a sealed bottle aside.
3.5 neutral alumina SPE. 1000mg, 3mL.
3.6 C18 solid phase extraction column. 1000mg, 6mL.
3.7 Organic filter. 0.45μm.
3.8 avermectin standard substance. (Abamectin, C48H72O14, CASN O. 71751-41-2), avermectin Bla content greater than 87%
The following avermectin content are avermectin Bla meter.
3.9 avermectin standard stock solution. accurately weighed amount (accurate to 0.0001g) avermectin standard substance dissolved in acetonitrile concentration formulation
As the standard stock solution 100μg/mL, and kept in -18 ℃ refrigerator.
3.10 avermectin intermediate standard solution. Pipette accurately avermectin stock standard solution, dilute with acetonitrile formulated marked as containing 1μg/mL concentration
Quasi intermediate liquid. Stored at 4 ℃ refrigerator.
3.11 avermectin standard working solution. accurate Pipette the appropriate amount of avermectin intermediate standard solution as necessary to set the volume of acetonitrile and diluted to the appropriate concentration
Standard working solution degrees. Stored at 4 ℃ refrigerator.4 instruments and equipment4.1 High performance liquid chromatography - mass spectrometry/mass spectrometry with atmospheric pressure chemical ionization (APCI source).
4.2 homogenizer.
4.3 centrifuge, 3000r/min or more.
4.4 Vortex.
4.5 Solid phase extraction device.
4.6 rotary evaporator.
4.7 nitrogen blowing instrument.
