3 Method summary
Latex protein in medical gloves and natural rubber products can be extracted from the buffer solution and precipitated to remove soluble interferences.
The Lowry method measures the absorbance at a wavelength of 750 nm from a spectrophotometer.
4 Terms and definitions
The following terms and definitions apply to this standard.
4.1
The sodium hydroxide capacity (mL) used to dissolve the sample to extract the precipitate and the sodium hydroxide used to decompose the standard ovalbumin extract
The ratio of capacity (mL).
Note. For example, 5mL test extract, precipitated and dissolved with 1.25mL sodium hydroxide, standard egg protein is dissolved with sodium hydroxide, and its dissolved volume is also
At 1.25 mL, the dilution rate of the test extract is the same as the dilution rate of the standard curve.
4.2
Latex proteins and polypeptides from natural rubber and its products.
4.3
Quantitative detection limit LOQ
The lowest protein content that can be quantitatively measured and produces accurate results. The LOQ is set to a standard deviation of 10 times.
5 instruments
5.1 Centrifuge.
5.2 Spectrophotometer.
5.3 Vortex mixer.
5.4 Micro-adjustable pipette (0.2mL ~ 5.0mL), volumetric flask (50mL, 100mL).
5.5 Polypropylene test tubes. capacities of 50 mL and 10 mL.
