Paralytic shellfish toxin in imported and exported shellfish products
Test method for high performance liquid chromatography
1 Scope
This standard specifies the sampling, sample preparation and liquid chromatography detection methods for the detection of paralytic shellfish toxins in imported and exported shellfish products.
This standard applies to the inspection of paralytic shellfish toxins in imported and exported shellfish products.
2 sampling and sample preparation
2.1 Inspection lot
No more than 10,000 pieces for an inspection lot.
Goods in the same inspection lot should have the same characteristics, such as packaging, labeling, origin, specifications and grades.
2.2 Number of samples
See Table 1.
Table 1 Sample quantity
Batch/piece
Frozen product, salt collection
Minimum number of samples/piece
150 and below 90 and below 3
151~3200 91~500 5
3201~1000 501~1200 13
- 1201~10000 20
2.3 Sampling method
The number of samples according to 2.2 is randomly selected and opened one by one. Take 500g from each piece as the original sample, the total amount of the original sample is quite a lot.
At 4kg, put it in a clean container, seal it, and send it to the laboratory in time.
2.4 Preparation of samples
Take the edible portion from the original sample taken, rinse off the dirt and sediment with water, and carefully remove excess with filter paper or clean gauze.
The water was placed in a pulverizer and pulverized to a homogenate, and was divided into two portions and placed in a clean container as a sample. Seal and mark.
2.5 Sample storage
Store the sample at -18 ° C or less in the dark.
Note. During sampling and sample preparation, samples must be protected from contamination or changes in residue levels.
3 Determination method
3.1 Method summary
The paralyzed shellfish toxin in the sample is extracted with 0.1 mol/L hydrochloric acid. After centrifugation, the supernatant is purified by C18 solid phase extraction column, and then purified.
Filtered through a 10000D ultrafiltration centrifuge tube, and the filtrate is separated by high performance liquid chromatography. After the column is derivatized, the fluorescence is detected.
Measurement, external standard method quantitative.
3.2 Reagents and materials
Unless otherwise specified, the reagents used were of analytical grade and the water was deionized water.
3.2.1 Acetonitrile. chromatographically pure.
