Foreword
This standard was drafted in accordance with GB/T 1.1-2009 given rules.
Instead of the standard SN/T 1146-2002 "tobacco ringspot virus quarantine and identification methods."
This standard compared with SN/T 1146-2002, the main technical changes are as follows.
--- Quarantine sampling site according to SN/T 2122-2008 "entry and exit quarantine of plants and plant products sampling" execution (see 6.2);
--- Remove the original standards Appendix A;
--- Added "symptom checker" one (see 6.3);
--- Immune electron microscopy method according to SN/T 1840-2006 "plant virus immune electron microscopy detection method" standard (see 7.3).
This standard is proposed and managed by the National Certification and Accreditation Administration Committee.
This standard was drafted. People's Republic of China, Shenzhen CIQ.
The main drafters of this standard. Chen Chi-nan, Tang Qiong, Cheng Yun Ling apricot, Zheng Wenhua, Wang Dongyong.
This standard replaces the standards previously issued as follows.

Tobacco ringspot virus quarantine and identification methods
1 Scope
This standard specifies the export plant quarantine of tobacco ringspot virus in quarantine and identification methods.
This standard applies to the entry and exit of plants and their products in quarantine and identification of tobacco ringspot virus.

Principle 3
Tobacco ringspot virus (tobaccoringspotvirus, referred TRSV), iso-called Anemone necrosis virus (anemonenecrosis
virus), blueberry necrotic ringspot virus (blueberrynecroticringspotvirus), tobacco ringspot virus No. 1 (tobaccoringspot
virus1). The virus is an isometric polyhedral spherical virus with a diameter of 25nm ~ 29nm, nematodes pass polyhedron virus is a member, can infect
54 Section 300 kinds of plants, the main cash crops are beans, potato, sweet potato, tobacco, watermelon, cucumbers, melons, carrots, Tangchang
Po, lily, iris, Prunus, apples, grapes, sweet cherries. Symptoms of plant infection by TRSV vary by host, usually appear blade ring
Or linear pattern, mottled or chlorotic spots, necrotic spots; root rot, stem dieback, are the fruit of malformations. The virus mainly rely on nematodes and seeds
Spread, mainly in North America, Oceania, Europe, Africa and other regions, with a broad host range, symptoms harm, immunogenicity, easy to make
Preparation of high titer, specifically the strong resistance of the virus antisera and other characteristics (see Appendix A). Serologically double antibody sandwich (DAS-ELISA) binding
Identification of host biological identification method or immune electron microscopy, as identified in the virus quarantine work basis.
4 utensils, equipment, facilities
Mortar, micro juicer, microplate reader, low-speed centrifuge (speed of 4000r/min), bio-incubator (with light, adjustable temperature range
Wai is 0 ℃ ~ 50 ℃), ELISA plate (48 or 96 wells), Eppendorf tubes (volume of 1.5mL), single channel pipette (10μL, 100
μL, 1000μL), 8-channel pipette (100μL or 200μL), heated water bath or incubator, analyze electronic balance, pH meter, white
