Foreword
This standard was drafted in accordance with the rules given in GB/T 1.1-2009.
This standard is proposed and managed by the Protection Department of the State Forestry Administration.
This standard was drafted. Sichuan University, Institute of Zoology, Chinese Academy of Sciences, China Giant Panda Conservation Research Center, Chengdu Giant Panda Breeding Research
Base, Sichuan Wildlife Resources Survey and Protection Management Station.
The main drafters of this standard. Yue Bisong, Wei Fuwen, Li Desheng, Shen Fujun, Yang Xuyu, Gu Xiaodong, Zhang Ling, Chen Hao.
Technical regulations for the establishment of genetic records of giant panda population
1 Scope
This standard specifies the collection and preservation of giant panda samples, DNA extraction, gender identification, individual identification, genetic diversity assessment and population inheritance.
The techniques and methods of archival establishment.
This standard applies to captive giant pandas and wild giant pandas.
2 Terms and definitions
The following terms and definitions apply to this document.
2.1
Microsatellite sequence microsatelitesequence
A DNA tandem repeat consisting of one to six nucleotides of a basic repeating unit in the genome, such as (AC)n, (ACG)n,
(ATTT)n, etc., also known as short tandem repeats (STR) or simple repeats (simplesequences)
Repeats, SSR). A polymorphic microsatellite sequence that changes in the number of repetitions between individuals can be used as a microsatellite marker.
2.2
DNA sample DNAsamples
Giant panda genomic DNA extracted from giant panda tissue (such as hair, blood, semen, muscle tissue, etc.) or fresh feces.
3 sampling frequency, time and route
3.1 sampling frequency and time
The collection of wild giant panda samples can be carried out in conjunction with daily work such as protected area monitoring and patrolling.
Large-scale centralized sampling of wild giant panda samples can be carried out after spring snow (breeding season) and winter snow accumulation (non-breeding season);
In the densely distributed area of giant pandas with convenient transportation, the number of sampling times can be increased according to specific conditions and work targets. All localities can be based on local
Giant panda's habits, as well as local phenology, climate, bamboo shoots of giant panda staples, field working conditions and logistical support, etc.
Volume sampling time. Field sampling should be completed in the shortest possible time, usually 10d~20d.
Captive giant panda samples can be collected at any time in conjunction with daily work.
3.2 Sampling route
The large-scale centralized collection route of the sample is based on the fourth giant panda survey method in the country, and the habitat of the giant panda in the reserve is divided into
Several grids with an area of 2km2, each grid as a sampling cell, and numbered. In a single cell, according to the topography and the road
The actual sampling route is laid out by factors such as roads, rivers, vegetation, and giant pandas.
The daily collection of samples can be determined based on the monitoring patrol route.
4 Sample collection and preservation methods
4.1 Sampling tools
Handheld terminal equipment, satellite locator, topographic map, record form, disposable PE gloves, 500mL plastic sample bottle, analytical pure water
