1 Scope

This standard specifies the MPN determination method for sulfate-reducing bacteria in seawater. This standard applies to the determination of sulfate-reducing bacteria in seawater and seawater utilization.

3 Terms and definitions

The following terms and definitions apply to this document. 3.1 Sulfate-reducing bacteria A type of bacteria that can reduce soluble sulfate in water to hydrogen sulfide under anaerobic or anoxic conditions. 3.2 MPN method MPN (mostprobablenumber) method Multi-tube fermentation technology is used to estimate bacterial concentration using probability theory. 3.3 Agedseawater It is taken from the unpolluted sea area and stored in a dark place with sea water for more than 15 days after being put into a glass bottle.

4 Principle of the method

This method uses multi-tube fermentation technology, cultured at (29 ± 1) ℃ for 21 days, if the bottom of the test tube produces black iron sulfide precipitate with sulfide Hydrogen odor indicates a positive reaction. The MPN method is used to count the sulfate-reducing bacteria in the test sample.

5 Reagents and materials

In this test method, except for special regulations, analytical reagents and water that meet the third grade water specifications in GB/T 6682 should be used.
