1 Scope

This document describes a method for the determination of capsanthin in feeds by HPLC. This document applies to capsanthin (the sum of capsanthin and capsorubin) in compound feed, concentrated feed and vitamin premix feed. 4 Principle

Capsanthin and capsanthorubin in the sample were extracted with acetonitrile and determined by high performance liquid chromatography and quantified by external standard method.

5 Reagents or materials

Unless otherwise specified, only analytical grade reagents were used. 5.1 Water. GB/T 6682, Grade 1. 5.2 Acetonitrile. chromatographic grade. 5.3 Phosphoric acid solution (0.3%, volume fraction). Pipette 1.5 mL of phosphoric acid, dilute to 500 mL with water, and mix well. 5.4 Capsanthin and capsanthin standard stock solution (100 μg/mL). weigh capsanthin (CAS No.. 465-42-9, purity ≥ 95%) and capsanthin (CAS No.. 470-38-2, purity ≥ 95%) standard products 10 mg each (accurate to 0.01 mg) were placed in Place in a 100 mL brown volumetric flask, dissolve with acetonitrile (5.2), make up to volume, and mix well.
