1 Scope
This document describes a HPLC method for the determination of neomethylhesperidin dihydrochalcone in feed.
This document applies to the use of neomethyl hesperidin dihydrochalcone in compound feed, concentrated feed, concentrate supplement and compound premix feed.
Determination.
The detection limit of this method in compound feed, concentrated feed and concentrate supplement is 0.5mg/kg, and the quantification limit is 1.0mg/kg;
The detection limit of premixed feed is 2.5 mg/kg and the quantification limit is 5.0 mg/kg.

4 Principle
The neomethyl hesperidin dihydrochalcone in the sample was extracted with methanol-formic acid acetonitrile solution and purified by solid phase extraction column.
The content was measured by spectrometer and quantified by external standard method.
5 Reagents or materials
Unless otherwise specified, only analytical grade reagents were used.
5.1 Water. GB/T 6682, Grade 1.
5.2 Methanol. chromatographic grade.
5.3 Formic acid. chromatographic grade.
5.4 Acetonitrile. chromatographic grade.
5.5 Methanol solution (90%, volume fraction). Take 90 mL of methanol (5.2), dilute to 100 mL with water, and mix well.
5.6 Formic acid-acetonitrile solution (2%, volume fraction). Pipette 2.0 mL of formic acid (5.3), dilute to 100 mL with acetonitrile (5.4), and mix well.
5.7 Extraction solution. Take 400 mL of methanol (5.2) and 100 mL of formic acid-acetonitrile solution (5.6) and mix well.
5.8 Formic acid solution (0.1%, volume fraction). Pipette 1.0 mL of formic acid (5.3), dilute to 1000 mL with water, and mix well.
