1 Scope
This standard specifies the principle, reagents and materials, instruments and equipment for the determination of water-soluble dietary fiber of cereals and their products by enzyme gravimetric method.

3 Principle
The cereal and product samples are hydrolyzed under buffer solution and high temperature and high pressure, and the sample is gelled by heat-stabilized α-amylase, and filtered.
The sample is digested by protease and amyloglucosidase to remove protein and starch, and the residue can be weighed by indigestible residue to remove ash.

4 reagents and materials
Unless otherwise stated, the reagents used shall be of analytical grade and the test water shall comply with the requirements for secondary water in GB/T 6682.
4.1 Anhydrous sodium acetate (CH3CO2Na).
4.2 Anhydrous sodium hydrogen phosphate (Na2HPO4).
4.3 Sodium dihydrogen phosphate (NaH2PO4·H2O).
4.4 Acetic acid (CH3CO2H2).
4.5 Anhydrous ethanol (CH3CH2OH).
4.6 Acetone (CH3COCH3).
4.7 Sodium hydroxide (NaOH).
4.8 Hydrochloric acid (HCl).
4.9 Protease.
4.10 Alpha-amylase (type VI-B extracted from pig pancreas).
4.11 Amyloglucosidase.
4.12 Glass wool.
4.13 2.0 mol/L buffer.
4.13.1 Acetic acid solution (2.0mol/L). Take 115mL of acetic acid (4.4), add 700mL of water, mix and dilute to 1L with water.
