Appendix A
(Normative appendix)
Determination of glutaraldehyde content
A.1 Determination of glutaraldehyde (C5H8O2) content
A.1.1 Principle
The glutaraldehyde reacts with the triethanolamine solution, and the neutral solution of hydroxylamine hydrochloride is used as the indicator, and the remaining triethanolamine solution is titrated with sulfuric acid standard solution.
Solution, calculate the content of glutaraldehyde based on the amount of sulfuric acid standard solution.
A.1.2 Reagent preparation
65g/L triethanolamine solution, 1% hydrochloric acid solution, 10g/L sodium hydroxide solution, 0.4g/L bromophenol blue ethanol solution and hydroxylamine hydrochloride
Solution (17.5g hydroxylamine hydrochloride added 75mL distilled water to dissolve, and add isopropanol to dilute to 500mL, shake well. Add 0.4g/L bromophenol blue ethyl
15mL of alcohol solution, titrated with 65g/L triethanolamine solution until the solution shows blue-green). Prepare and calibrate 0.25mol/L sulfuric acid titrant
(See A.2).
A.1.3 Sample determination
Precisely draw an appropriate amount of the sample to make it equivalent to about 0.2g of glutaraldehyde, put it in a 250mL iodine flask, and accurately add 65g/L triethanolamine solution
20.0mL and 25mL of hydroxylamine hydrochloride neutral solution, shake well. After standing the reaction for 1h, use 0.25mol/L sulfuric acid titrant (installed in 25mL
In the burette) titration. When the solution turns blue-green, record the amount of sulfuric acid titrant. At the same time, use glutaraldehyde-free triethanolamine and hydroxylamine hydrochloride
Repeat the above operation for the solution (blank control). Repeat the measurement twice, and take the average of the two times for the following calculations.
A.1.4 Calculation formula
Since 1mL of 1mol/L sulfuric acid titrant is equivalent to 0.1001g of glutaraldehyde, the glutaraldehyde content is calculated according to formula (A.1).
A.1.5 Matters needing attention
This method is suitable for determining the content of glutaraldehyde in aldehyde disinfectants. The key to the method is to prepare a neutral solution of hydroxylamine hydrochloride
At the time, the blue-green should be spotted. For high-concentration glutaraldehyde, it should be pre-diluted to a dilute solution with a mass fraction of about 2% before measurement. Correct
For alkaline or acidic glutaraldehyde samples, first adjust the pH to 7.0 with 1% hydrochloric acid or 10g/L sodium hydroxide solution, and then use the above method for content determination.
A.2 Sulfuric acid (H2SO4) titrant
A.2.1 Titration solution preparation
When preparing 0.25mol/L sulfuric acid titrant, take 15mL of sulfuric acid and slowly pour it into water along the wall of the beaker filled with distilled water. Wait for the solution temperature
When the temperature drops to room temperature, add distilled water to dilute to 1000mL, and shake well.
A.2.2 Concentration calibration
When calibrating the concentration, weigh the reference anhydrous sodium carbonate 0.4g (accurate to 0.0001g) dried to constant weight at 270℃~300℃, and set
In a 250mL iodine measuring flask, add 50mL of distilled water to dissolve. Add methyl red-bromocresol green mixed indicator solution (1g/L methyl red ethanol solution
Mix 20 mL with 30 mL of 2g/L bromocresol green ethanol solution) 10 drops, use the prepared sulfuric acid titrant (put into a 50mL burette)
set. When the solution turns from green to purple-red, boil for 2 minutes. After cooling to room temperature, continue to titrate until the solution changes from green to dark purple.
Record the total milliliters of sulfuric acid titrant used.
A.2.3 Concentration calculation
Because 1mL of 1mol/L sulfuric acid titrant is equivalent to 0.1060g of anhydrous sodium carbonate, the concentration of sulfuric acid titrant is calculated according to formula (A.2).
