Including field survey, indoor testing, issuing certificates and supervise the production units to do the election, the selection and disease treatment and so on.
2.2
Plants, plant products or other regulated articles official visual inspection to determine if pests are present, compliance with phytosanitary
Regulations.
2.3
To determine if pests are present or in addition to the visual inspection for the official identification of the pest carried out.
2.4
For areas threatened by its potential economic importance, but not in place in the region, or, having incurred but not widely distributed and official proof

6.2.1 Sample collection
Collection show symptoms suspicious diseased plant (block), or select growing weak, yellow, small, wilted plants, soil collected in and around the tubers,
Back to the laboratory for indoor testing; No suspicious diseased plants (blocks) of the sample collection part of the sample plots indoor testing. Fill out the "pest
Survey Sampling was record "(see Appendix E).
6.2.2 Detection
Quarantine pest detection method carried out according to the relevant standards; no standard, according to conventional methods for testing. Stem rot nematode
Detection See Appendix F.
6.2.3 results found
The test results filled with "harmful biological sample appraisal report" (see Appendix G). If necessary, to prepare specimens to be saved.
7 disease treatment
7.1 Seed found with quarantine pests, immediately stop breeding.
7.2 outbreak occurred seedlings, quarantine fumigation treatment or take place burned, digging deep and other measures. Not yet show symptoms of potato
Block, the need for quarantine fumigation treatment can not quarantine quarantine treatment may be made after concentrated feed processing cooked.
7.3 occurred quarantine pests and disease to disease to surrounding plots, should replant the pest non-host crops.
8 Visas
8.1 After field, indoor inspection found no quarantine pests of seedlings, plant quarantine agencies to issue "quarantine certificate" (see Annex
Record H). "Quarantine certificate" valid for 1 year.
8.2 found quarantine pests, quarantine insecticide treatment shall be issued "quarantine certificate"; quarantine insecticide treatment failure
And not the issue of "quarantine certificate" and inform the applicant or quarantine individuals.

Field stem rot nematode symptoms
C. 1 seedling symptoms
Seedbed emergence little yellow dwarf, the white part of the stem base seedlings appear mottled, after changes to black, there is a gap in the stem cut open, brown or pith
Purple, broken little stream does not flow or white pulp.
C. 2 Field Period Symptoms
Early symptoms of growth, seedling vine short mid performance, near-surface cracks potato Shui epidermis, leaf yellowing from the bottom up, these disorders
Like the victim is caused by the underground tubers.
C. Tuber symptoms on 3
There are three common types. One is bran heart-shaped, from the onset of tuber size, color and other aspects of normal potato no significant difference between the epidermis
Intact, but the interior due to the tubers after stimulation by the nematode, thin-walled cell dehydration, shrinkage white spongy (or powder), a large gap, called
"Bran Heart." Since the latter part of the soil bacteria opportunistic infections, therefore rendering brown and white heart-shaped bran (severe skin was dark brown - liver
color). This type is more direct infection caused by the nematode with seedlings; the other is split skin type, tuber skin cracking, dehydration shrinkage. This class
Type directly from the soil nematode infestation caused by sucking; third type is mixed, the performance of the internal hollowing, crack external skin.

Stem rot nematode indoor test methods
F. Sample 1
In potato breeding period, we found seedbed sparse seedlings growing weak, even rotten yellow dwarf seedlings showing symptoms should check each tree seedlings, seedling disease and will be
Rhizosphere soil taken back to the laboratory for nematode Isolation and identification. During the field, in the sweet potatoes and cut dry matter yield planing, the disease is the best time of the survey
Of collecting tubers showing symptoms, back to the laboratory for nematode Isolation and identification.
F. Save 2 samples
The collected samples back to the lab separated in time, if not separated in time, samples can be stored in a refrigerator 4 ℃ ~ 10 ℃ in.
F. 3 separate samples nematodes
F. 3.1 Baermann funnel method. selection of the diameter of 10cm ~ 14cm glass funnel, and then the following section of latex tubing, latex tube tops a seal
Clip, filled with water in the funnel, the funnel placed on the shelves, the chopped plant material or soil sample with a layer 2 to 3 layers of gauze or high toughness
Grade tissue wrap gently on the funnel, after 24h ~ 48h, nematodes due to its water-based and tend to sink under its own weight of milk under the hopper
The hose with a small petri dish or test tube bonding under latex tube, release the clip seal collected nematode suspension.
F. 3.2 platter funnel method. the sample is placed covered with 2 layers of gauze or tissue of small sieve tray, and then put into a small sieve tray filled with water funnel
The other procedures were the same Berman funnel method. Small sieve tray diameter smaller than the diameter of the funnel 2cm ~ 3cm, depth of 2cm, mesh diameter of 0.2cm ~
0.5cm.
F. 3.3 shallow basin method. The sample was laid flat on a 2-layer gauze or tissue sieve plate, sieve bowl on the amount of water containing the bottom of the basin,
Just soak in water samples is appropriate, after 24h ~ 48h, remove the bowl screen, the bottom of the pot nematode suspension was passed through a 400 mesh (pore size 38μm) of
Sieve collected nematodes.
F. 4 examination
Separation of the resultant suspension of nematodes in a test tube and placed in water bath 60 ℃ ~ 65 ℃ in 2min ~ 3min kill nematodes. Already
Kill nematodes promptly fixed with 4% formaldehyde, made into slides under the microscope morphological nematode specimens were observed and measured, and the rot
Morphological characteristics rotten stem nematode comparison, if the match is determined nematode identified as D. destructor.
F. 5 Morphology
F. 5.1 Form
Female. parasites linear, hot kill parasites bent slightly to the ventral, lateral line 6. Head flat, slightly constricted, obviously stylet base ball, in
Esophagus ball spindle with flap, back covering after esophageal intestinal glands short (and occasionally constricted). Single ovary, reach, sometimes extends up to the esophagus area, after Yin child
Palace anal sac length is from 40% to 98% of the female pitch. Tail cone, usually curved belly, side round.
Male. the front and tail shape of the female body shape. Bursa extends 50% to 90% of the tail, spicules length 24μm ~ 27μm.
Female. L = 0.69mm ~ 1.89mm; Rao = 18 to 49; Chou = 4 to 12; Ba = 14 to 20; Ba '= 3-5; Vp = 77 to 84; mouth of the needle length =
10μm ~ 13μm.
Male. L = 0.63mm ~ 1.35mm; Rao = 24 to 50; Chou = 4 to 11; Ba = 11 to 21; stylet length = 10μm ~ 12μm.
Major surveying projects (DeMan formula).
L --- body length;
Rao --- length/maximum body width;
Chou --- Length/distance from the front end of the esophagus to the end;
Vp front end --- the distance vulva × 100/body length;
Ba --- body length/tail length;
Ba '--- tail length/width anus body.
F. 6 equipment required
Microscope 1; dissecting microscope 1 set; funnel 1; Iron frame 1; a shallow dish; sieve group; a bottom basin; latex tube 1; seal
Clip one; several tubes; a plurality of dish; gauze 2; pick a needle.
F. 7 fixative formulation
Standard formaldehyde fixative. Formalin (40% formaldehyde). distilled water = 1.9.
Double glycerol formaldehyde fixative. Formalin (40% formaldehyde). distilled water = 2.8.
