1 Scope
This standard specifies the residues in imported and exported food Jinggangmeisu liquid chromatography - mass spectrometry/mass spectrometry methods.
This standard applies to rice, cabbage, onions, carrots, tomatoes, cucumbers, spinach, mushrooms, pears, lemons, almonds, tea, pork, liver, Romania
Non-fish, shrimps Jinggangmeisu residue detection and confirmation.
2 Method summary
The sample Jinggangmeisu residue extracted with aqueous methanol by HLB solid phase extraction column or liquid-liquid extraction with ethyl acetate purified by liquid
Chromatography - mass spectrometry/mass spectrometry detection and confirmation, external standard.
3 Reagents and materials
Unless otherwise stated, all reagents HPLC grade water is deionized water.
3.1 ammonium acetate.
3.2 methanol.
3.3 acetonitrile.
3.4 ethyl acetate.
3.5 glacial acetic acid.
3.6 methanol + water (9 + 1, volume ratio). Measure 90mL of methanol and 10mL of water, and mix.
3.7 5mmol/L ammonium acetate buffer solution (pH4.5). Weigh 0.385g ammonium acetate dissolved in water and dilute to 1000mL, ice B
Acid to adjust the pH to 4.5 ± 0.1.
3.8 Standard substance. Jinggangmeisu A (validamycinA, CAS. 37248-47-8), molecular formula. C20H35NO13, purity 91.0%.
3.9 standard stock solution. Weigh 2mg (accurate to 0.1mg) Jinggangmeisu standard product, dissolved in water and dilute to 25mL brown capacity
Bottle, formulated at a concentration of 80mg/L of standard stock solution.
3.10 standard solution matrix. if necessary, with a blank sample extract obtained in accordance with the sample preparation step, preparation of different concentrations of base

3.11 HLB column. 6mL, 200mg or equivalent person. Before using successively activating 6mL methanol and 6mL water.
3.12 0.22μm membrane, bipolar.
4 instruments and equipment
4.1 liquid chromatography tandem quadrupole mass spectrometer. with atmospheric pressure chemical ionization (APCI).
