1 Scope

4 Equipment and Materials
4.1 incubator. 36 ℃ ± 1 ℃.

5 medium and reagents
5.1 sterile saline. Weigh 8.5g of sodium chloride dissolved in 1000mL of distilled water, 121 ℃ autoclave 15min.
5.2 1mol/L sodium hydroxide (NaOH). Weigh 40g of sodium hydroxide (NaOH) was dissolved in 1000mL of distilled water.
5.3 1mol/L hydrochloric acid (HCl). Pipette concentrated hydrochloric acid 90mL, diluted with distilled water to 1000mL.
