DNA barcode species identification procedures
1 Scope
This standard specifies the identification process of the quarantine pest DNA barcode identification system in China.
This standard is applicable to the identification of DNA barcode species in animal, plant and port health and quarantine pests in China.
2 Terms and definitions
The following terms and definitions apply to this document.
2.1
DNA barcode technology DNAbarcoding
Use standardized, sufficiently variable, easily amplified and relatively short gene fragments for species identification and new species discovery
Identification technology.

2.3
S value and E value S-scoreandE-value
The S value (S-score) indicates the degree of similarity between two DNA sequences (query sequence and target sequence), and the higher the score indicates the phase between them.
The greater the degree. The E value (E-value) is an evaluation of the reliability of the S value. The E value indicates other target sequences and queries in a random case.
The similarity of the sequence is higher than the likelihood of the similarity between the query sequence and the target sequence. The lower the E value, the more reliable the S value, that is, the target sequence
The higher the similarity with the query sequence. When the E value is less than 10-5, it indicates that the two sequences have high similarity.
2.4
FASTA format FASTAformat
In bioinformatics, a format based on text used to represent nucleotide sequences or amino acid sequences. Base pair in this format
Or amino acids are encoded in a single letter and allow the addition of sequence names and annotations before the sequence. The first line of the sequence file is made up of the greater than sign " >"
Start with a sequence tag. Starting from the second line, the sequence information is only allowed to use the given nucleotide or amino acid coding symbols. Usually nuclear
The nucleotide symbol can be capitalized, while the amino acid is only in uppercase letters.
Note. See Appendix A for an example.
2.5
DNA barcode DNAbarcodes
CO1. mitochondrial cytochrome c oxidase subunit 1, animal DNA barcode
ITS. internal transcribed spacer, fungal and plant DNA barcode
rbcL. chloroplast gene, plant DNA barcode
matK. chloroplast gene, plant DNA barcode
16S. 16S ribosomal RNA, bacterial DNA barcode
