Chemicals - Bovine corneal opacity and permeability test People's Republic of China Entry - Exit Inspection and Quarantine Industry Standard Chemical corneal opacity and permeability test Released by.2015-02-09 2015-09-01 implementation People's Republic of China State Administration of Quality Supervision, Inspection and Quarantine issued

Foreword

This standard is equivalent to the United Nations Economic Cooperation and Development Organization (OECD) Chemical Test Method 437 "Bovine corneal opacity and permeability test Test "(2009). This standard differs from the OECD Chemical Test Method No.437, with the following differences. --- Editorial changes to OECD Chemical Test Method No.411; --- Added preface. This standard is proposed by the State Administration of Quality Supervision, Inspection and Quarantine. This standard is under the jurisdiction of the National Certification and Accreditation Administration. The drafting unit of this standard. Tianjin Entry - Exit Inspection and Quarantine Bureau of the People's Republic of China. The main drafters of this standard. Zhao Zhuo, Liu Ming, Liu Fengjuan, Feng Jie, Zhang Bin, Wang Na, Yu Yanyan. Chemical corneal opacity and permeability test

1 Scope

This standard specifies test preparation, test methods and results reports for bovine corneal opacity and permeability testing (BCOP). This standard applies to chemical eye corrosive or severe eye injury identification test.

2 normative reference documents

The following documents are indispensable for the application of this document. For dated references, only the dated edition applies to this article Pieces. For undated references, the latest edition (including all modifications) applies to this document. GB/T 21609 Chemical acute eye irritation/corrosion test method

3 terms and definitions

The following terms and definitions apply to this document. 3.1 Cornea cornea The transparent part of the front of the eye, covering the pupil and iris, and can receive light. 3.2 Corneal opacity cornealopacity A method of detecting the opacity of the cornea after exposure to a test substance. An increase in corneal opacity indicates that it is damaged. Can follow GB/T 21609 in the method of subjective evaluation of corneal opacity, but also through the instrument to its objective evaluation, such as "turbidity meter." 3.3 Corneal permeability. Cornealpermeability Quantitative analysis of corneal endothelium damage was performed by measuring the amount of fluorescein sodium dye passing through the corneal cell layer. 3.4 In vitro Stimulation Score invitroirritancyscore (IVIS) Using the empirical formula derived from the BCOP experiment, this formula integrates the average turbidity and average permeability of each treatment group into Each group scored separately in vitro. IVIS = average turbidity value (15 x average permeability). 3.5 Turbidimeter opacitometer An instrument that calculates "corneal opacity" by quantitatively analyzing the transmitted light from the cornea. The standard instrument consists of two parts, each of which Each has its own light source and photoelectric tube, one for the treated cornea and the other for instrument calibration and zeroing. Halogen light Of the light can be controlled by the control components (empty and window empty containers) to the photoelectric tube, but also through the experimental components (container containing cornea) to reach the light Electric tube. Analyze and compare the difference in light transmittance between the two types of photoelectrics, calculate the turbidity values and display them in digital form (see Appendix B).

4 test preparation

4.1 Bovine eye source and requirements Obtain cattle from the regular slaughterhouse. Cattle eye should be taken from cows larger than 12 months of age and less than 60 months of age.
