1 Scope
This standard specifies the method for inspection and identification peach rosette mosaic virus.
This standard applies to quarantine and identification of peach, grapes and other plant material peach rosette mosaic virus.

3 equipment, appliances, and reagents
3.1 Equipment
ELISA detector, real-time PCR instrument, low temperature refrigerators, incubators, autoclaves, electronic analytical balance, high-speed desktop centrifuged
Machine, constant temperature water bath, ice-making machines.
3.2 Appliances
Micro adjustable pipette (range, respectively 2.5μL, 10μL, 100μL, 200μL and 1mL) and sterilized tip, 1.5mL centrifugation
Tube, mortar, ELISA plate, 0.2mLPCR tubes, 96-well PCR reaction plates and lids.
3.3 Reagents
Reagents, buffers, primers and probe sequences, etc. See Appendix B and Appendix C.
4 Detection and Identification
4.1 Serology (double antibody sandwich enzyme-linked immune assay)
4.1.1 Coating antibody
The PRMV antibody with the coating buffer was diluted to working concentration, enzyme-linked plate, each hole by adding 100μL. The ELISA plate stamped
(Or sealed with plastic wrap) into the humidor, placed in 37 ℃ incubator incubated for 2h (or 4 ℃ refrigerator overnight). After each well was added
300μLPBST washed three times, each time 3min.
4.1.2 Sample Preparation
Plant leaves to be tested was weighed 0.2g, was added 2mL sample extraction buffer, followed by sufficient grinding, into a 1.5mL centrifuge tube,
5000r/min centrifugal 5min, the supernatant 1.10 dilution of the sample is a crude extract.
4.1.3 sample
The sample was added to the crude extract of enzyme-linked antibody-coated plates, each well 100μL. Konga buffer as control, at the same time set
