1 Scope
This standard specifies the total number of colonies, coliforms, fecal coliforms and E. coli in the import and export beverages.
Membrane method.
This standard is applicable to the determination of drinking and drinking natural mineral water, bottle (barrel) drinking pure water, tea beverage and carbonated beverage.
2 Method summary
When a certain amount of sample liquid passes through the filter membrane, the bacteria are trapped in the square of the filter membrane, and after the filter membrane is cultured on the medium, the bacteria are counted.
The number of colonies can be measured to determine the total number of colonies; if the filter is placed on selective medium, the positive colonies will be colored in a specific color, counting
The number of positive colonies in these specific colors can be used to determine the number of positive colonies in the sample.

4.10 Filter paper.
5 sample preparation
5.1 Samples with low bacterial content can be directly filtered using a sterile pipette with 50 mL.
5.2 Based on an estimate of the contamination of the sample, the sample is made into a series of 10-fold increasing sample dilutions using sterile saline. preparation
The entire sample should not be allowed to exceed 15 min and then filtered with 50 mL of sample dilution.
