Foreword

This standard is drafted in accordance with the rules given in GB/T 1.1-2009. This standard replaces SN/T 0944-2000 "Indigo test method for export of frozen shrimp and shrimp products". Compared with SN/T 0944-2000, the main technical changes are as follows. --- modified standard name (see title). --- delete the preamble "This standard also developed a sampling and sample preparation method" (2000 edition of the preface); - change the range to "shrimp and shrimp" (see Chapter 1, Chapter 1 of the.2000 edition); - modified normative references (see Chapter 2, Chapter 2 of the.2000 edition); - the definition, sampling and sample preparation (Chapter 3, Chapter 4 of the.2000 edition); --- increase the second method of high performance liquid chromatography. Please note that some of the contents of this document may involve patents. The issuer of this document does not assume responsibility for the identification of these patents. This standard is proposed and approved by the National Certification and Accreditation Administration Committee. The drafting of this standard. Zhejiang Province Inspection and Quarantine Science and Technology Research Institute. The main drafters of this standard. Hou Jianbo, Xie Wen, Lu Chunhua, Lu Shun, Shi Yingzhu, Li Jie, Zhu Zilong. This standard replaced the previous version of the standard release. --- SN/T 0944-2000. Determination of Indole Content in Export Shrimp and Shrimp

1 Scope

This standard specifies the shrimp and shrimp dry indole content of spectrophotometry and high performance liquid chromatography detection method. This standard applies to the determination of indole content in shrimp and shrimp.

2 normative reference documents

The following documents are indispensable for the application of this document. For dated references, only the dated edition applies to this article Pieces. For undated references, the latest edition (including all modifications) applies to this document. GB/T 6682 Analytical laboratory water specifications and experimental methods First method spectrophotometric method

3 principle

The indole in the sample was distilled off by steam distillation, extracted with chloroform, and the developer was shaken. The acid layer was separated, Spectrophotometer determination, standard curve method quantitative.

4 reagents and materials

Unless otherwise specified, all reagents are of analytical grade and water is a Grade I water in accordance with GB/T 6682. 4.1 anhydrous ethanol. 4.2 Phosphoric acid (H3PO4). 4.3 concentrated hydrochloric acid (HCl). 4.4 Defoamer. Polyether. 4.5 saturated sodium sulfate. 4.6 Trichloromethane (CHCl3). 4.7 Reagent. p-Dimethylaminobenzaldehyde, purity greater than or equal to 98.0%. Preparation of the developer. Dissolve 0.4 g of p-dimethylaminobenzaldehyde in 5 mL of acetic acid, add 92 mL of phosphoric acid and 3 mL of hydrochloric acid. Since the purity of p-dimethylaminobenzaldehyde affects the strength of the reagent blank, if the reagent is yellow, it is purified as follows. To dissolve 100 g of p-dimethylaminobenzaldehyde in 600 mL of hydrochloric acid (16), add 300 mL of water, slowly stirring with stirring 10% sodium hydroxide solution precipitation of dimethylaminobenzaldehyde, a white precipitate, stop adding sodium hydroxide solution, filter, discard the precipitate. Following Continued and to p-dimethylaminobenzaldehyde mostly precipitated. Discard 4g ~ 5g color of p-dimethylaminobenzaldehyde precipitation. Remaining white The precipitate is filtered and washed with water to precipitate until the lotion is no more acidic. The p-dimethylaminobenzaldehyde is dried and should be white and placed in a desiccator In the preservation. 4.8 Acetic acid (CH3COOH). If the reagent reacts with the developer and becomes pink, it is purified by adding 500 mL of acetic acid, 25 g of potassium permanganate and 20 mL of sulfuric acid were added successively to a 1000 mL round bottom conical flask and distilled in an all-glass distiller to collect distillate Liquid should not be more than 400mL.
