Foreword

This standard was drafted in accordance with the rules given in GB/T 1.1-2009. This standard was proposed by the National Grain and Material Reserve Bureau. This standard is under the jurisdiction of the National Grain and Oil Standardization Technical Committee (SAT/TC270). This standard was drafted by. National Academy of Food and Material Reserve Research Institute, Guangdong Food Science Research Institute, Shandong Grain and Oil Testing Xin, Shaanxi Grain and Oil Product Quality Supervision and Inspection Center, China National Grain Management Group Co., Ltd., Beijing Grain, Oil and Food Inspection Institute, Heilongjiang Provincial Grain Quality and Safety Monitoring Center, Anhui Province Grain and Oil Products Quality Supervision and Inspection Station, Inner Mongolia Autonomous Region Grain and Oil Quality Testing Center, Hubei Province Grain, Oil and Food Quality Supervision and Inspection Center, Shanxi Grain Quality Monitoring Center, Henan Provincial Grain Science Research Institute Co., Ltd., Beijing Zhongjian Baotai Biotechnology Co., Ltd. Grain and oil inspection of organic phosphorus and Rapid qualitative detection of carbamate pesticide residues

1 Scope

This standard specifies the principles, reagents and reagents for rapid qualitative detection of organophosphorus and carbamate pesticide residues in grains and their products Materials, instruments and equipment, sample preparation and result analysis. This standard applies to the rapid qualitative detection of organophosphorus and carbamate pesticide residues in grains and their products. The method detection limit of this standard is shown in Appendix A.

2 Normative references

The following standards are essential for the application of this document. For dated references, only the dated version applies to this article Pieces. For the cited documents without date, the latest version (including all amendments) applies to this document. GB 2763 National Food Safety Standard Maximum Residue Limits of Pesticides in Food GB/T 5491 Grain and Oil Inspection Sampling and Sampling Method GB/T 6682 Analysis laboratory water specifications and test methods

3 Principle

Organophosphorus and carbamate pesticide residues in the sample inhibit the reaction of insect esterase and enzyme receptor, and the fluorescent substrate is activated to emit fluorescence The signal is weakened, and the enzyme inhibition rate of organophosphorus and carbamate pesticides in this sample is calculated by the change of the fluorescence signal and the corresponding limit Enzyme inhibition rate comparison, so as to quickly determine whether the organophosphorus and carbamate pesticides in the sample exceed the standard.

4 Reagents and materials

Unless otherwise specified, the reagents used in this method are analytically pure, and the water is secondary water specified in GB/T 6682. 4.1 Acetone. 4.2 Ethyl acetate. 4.3 Anhydrous sodium sulfate. 4.4 Ethyl acetate-acetone solution (95. 5, volume ratio). Take 5mL acetone and add 95mL ethyl acetate, mix well. 4.5 Sodium sulfate solution. Take.200g of anhydrous sodium sulfate, dissolve in water and dilute to 1000mL, mix well. 4.6 Phosphate buffer solution (hereinafter referred to as PBS). Weigh 8.00g sodium chloride, 1.20g disodium hydrogen phosphate (or 2.92g hydrogen phosphate dodecahydrate) Disodium), 0.20g potassium dihydrogen phosphate, 0.20g potassium chloride, dissolved in 900mL water, adjust the pH to 7.4 ± 0.1 with hydrochloric acid, and dilute with water to 1000mL. 4.7 1% TritonX-100 (or Tween-20) in PBS. take 10 mL TritonX-100 (or Tween-20), dilute with PBS to 1000mL.
