The environmental monitoring category that clarifies the environmental pollution situation.
3.2
Phytoplankton
It is a kind of autotrophic plankton, mostly single-celled plants, with chlorophyll or other pigment bodies, which can absorb light energy and
Carbon dioxide conducts photosynthesis and makes organisms (mainly carbohydrates) by itself. Mainly include diatoms, dinoflagellate, green
Algae, cyanobacteria, golden algae, xanthellae and algae spores, etc., they are the main producers of waters.
23.3
Zooplankton zooplankton
Small animals that live in the water layer and move passively are collectively called zooplankton. Including planktonic protozoa, coelenterate,
Pteropods and heteropods of molluscs, crustaceans, chaetognaths, tunicates, planktonic larvae and other phyla
Individual floating types, etc. According to individual size, it can be divided into giant zooplankton, large zooplankton, medium and small zooplankton and miniature zooplankton.
Zooplankton. The large zooplankton and medium and small zooplankton in this standard refer to the use of GB 17378.7 shallow water I and II respectively
Zooplankton collected by the type plankton net.
3.4
Microorganism
Refers to many groups of organisms that are small and generally require the help of a microscope to be recognized. Microorganisms in a broad sense include bacteria,
Actinomycetes, molds, yeasts, spirochetes, rickettsia, mycoplasma, chlamydia, viruses, viroids, protozoa
And unicellular algae.
3.5
Community structure
It is one of the characteristics of the overall level of the biological community, and the most important item in a series of attributes of the community. Community structure package
Including nutritional structure, spatial structure (vertical distribution and horizontal distribution), time structure (circadian rhythm and seasonal distribution) and physical
Kinds of structure and other aspects.
3.6
Dominant species
Refers to the species with a large number and high frequency in an ecosystem or community.
3.7
Species diversity
The index is measured by the number of species in a community and their relative abundance, including the number of existing species in the community
Order also includes the relative abundance of species (ie, homogeneity), that is, the abundance and heterogeneity of species within a community or ecosystem.
3.8
Evenness
An index reflecting the uniformity of the community structure.
3.9
Richness
An index indicating the abundance of species in a community (or sample).
3.10
Intertidal benthos
3 Plants living on the bottom surface of the intertidal zone and animals on the bottom surface and in the bottom.
3.11
Macrobenthos
The animals and plants that live on, in or near the bottom of waters are collectively called benthic organisms. Macrobenthos
A type of organisms, benthic organisms that cannot pass through a 0.5 mm mesh screen.
4 General requirements for biological monitoring in coastal waters
The development of the implementation plan for coastal biological monitoring shall be implemented in accordance with the relevant requirements of HJ 442.1; the layout of monitoring points shall be in accordance with HJ 730
Implementation requirements; data processing and data reporting shall be implemented in accordance with HJ 442.2; evaluation and reporting shall be implemented in accordance with HJ 442.10.
The biological monitoring frequency for routine monitoring of important estuaries and bays and key regions determined by the state is based on dry, high, and normal water periods each year.
Perform three times, sampling and water quality monitoring are combined; other areas can be carried out according to monitoring conditions and evaluation requirements, such as
Monitoring is carried out during high and normal water periods or the same water period is selected for monitoring once every 2 years.
Routine monitoring items include phytoplankton, macroplankton, chlorophyll a, fecal coliforms, macrobenthos (fixed
the amount).
The time and frequency of biological monitoring in other coastal waters are determined according to the monitoring purpose; other monitoring items generally include red tide
Animals, small and medium zooplankton, macrobenthos (qualitative), macroalgae, total number of bacteria and fish avoidance response.
5 Collection, storage and transportation of biological samples
5.1 Sampling level
a) The sampling level of each monitoring item shall be implemented in accordance with GB 17378.7 and HJ 442.3;
b) Microorganisms take surface samples;
c) The sampling level of chlorophyll a and phytoplankton quantitative samples is the same as that of water quality sampling, see HJ 442.3;
d) Qualitative phytoplankton samples and zooplankton samples are collected 2 m from the bottom and dragged vertically to the surface;
e) The collection of macrobenthos shall be implemented in accordance with GB 17378.7.
5.2 Sample sampling
a) Samples for each monitoring item are prepared and collected in accordance with GB 17378.7;
b) Preparation before sampling. According to the survey items, locations and levels, equip with sufficient sample bottles, fixatives and other devices
Use a suitable monitoring ship, the sampling operation must be carried out after the ship is stationary, according to the prevailing gas
The sampling position can be adjusted appropriately for elephant and ocean current conditions to ensure the convenience and safety of sampling;
c) Use sterile water collectors for microbial sampling, and pay attention to ensuring the aseptic operation of the entire process to avoid contamination;
d) Plankton sampling uses shallow water type Ⅰ, Ⅱ, and Ⅲ plankton nets, and the netting does not exceed 1 m/s; the netting is about 0.5 m/s;
e) Quantitative sampling of large benthic organisms uses a 0.1 m2 mud picker, 3 times at each point; in special circumstances, many
For 2 times; if conditions do not permit, a 0.05 m2 mud picker can be used, but the number of sampling needs to be increased; qualitative sampling
Use A's trawl, the trawling speed is controlled at about 2 knots, and the trawling time for each point is 10 min;
f) Quantitative sampling in intertidal zone. sampling in high, middle and low tide areas, quantitative sampling device for tidal flat quantitative sampling, sample square
The number is usually 8 per station (total 0.5 m2); if the beach sediment type is more uniform and the biological distribution is more uniform,
4 samples can be considered; quantitative sampling of rocky shore generally uses a 25 cm×25 cm quantitative frame, and 2 samples are taken from each station
Sample. If the habitat density of organisms is very high and the distribution is relatively uniform, a quantitative frame of 10 cm×10 cm can be considered;
g) Qualitative sampling in the intertidal zone. while sampling at each station, the species of animals and plants that appear near the station should be collected as much as possible.
The collection is complete for reference during analysis, but the qualitative samples must be separated from the quantitative samples and should not be confused.
5.3 Sample storage and method
Refer to GB 17378.7 for the fixation, storage and transportation of samples for monitoring items. The collection and storage of samples for monitoring items are often
See Table 1 for usage method.
a) Microbiological samples should be analyzed as soon as possible after collection, and the time should not exceed 2 hours, otherwise, the samples should be placed in an ice bottle or refrigerator
In, the time shall not exceed 24 h;
b) Chlorophyll a sample should be filtered immediately after collection, then wrap the filter membrane with aluminum foil, and dry it at -20℃
Dry and save for testing;
c) The phytoplankton water sample is fixed by adding 6 ml to 8 ml Lugol's solution per liter of water sample (Lugol's solution is iodine tablets dissolved in
Saturated solution prepared with 5% potassium iodide solution), shake well; add 5% formaldehyde solution to the net sample;
Add formaldehyde solution to 5% of the sample volume of the net sample and shake well;
d) After the zooplankton sample is collected, add a 5% formaldehyde solution with a volume fraction and shake well;
e) After collecting the macrobenthic samples, they are cleaned with seawater on site, and the volume fraction for temporary storage is 5%~7%
Neutral formaldehyde solution, 75% glycerol ethanol solution or 75% volume fraction for permanent storage
Ethanol fixation; if the fixed sample has not been separated and identified for more than two months, the fixative should be replaced once.
