1 Scope
This document describes the high performance liquid chromatography method for the determination of coumaric acid in bagasse.
This document is applicable to the qualitative and quantitative determination of coumaric acid in bagasse. Refer to the content detection of products containing coumaric acid

4 Principle
The bagasse sample is extracted with 2mol/L NaOH solution, the pH is adjusted to about 2.0 with 6mol/L HCl solution, and ethyl acetate is extracted.
After being taken, it was separated by C18 chromatographic column, detected by ultraviolet-visible light detector, chromatographic peak retention time was qualitative, and external standard method was used for quantification.
5 Reagents and materials
5.1 Reagents
Unless otherwise stated, all reagents are chromatographically pure, and the water is the first grade water specified in GB/T 6682.
5.1.1 Methanol (chromatographically pure).
5.1.2 Acetic acid (chromatographically pure).
5.1.3 Ethyl acetate (chromatographically pure).
5.1.4 Hydrochloric acid (high-grade pure 36%~38%).
5.1.5 Sodium hydroxide (analytical grade).
5.2 Reagent preparation
Acetic acid aqueous solution (0.1%). Pipette 0.25mL acetic acid (5.1.2) into a 250mL volumetric flask, and dilute to the mark with water.
