Food natamycin determination of liquid chromatography
1 Scope
This standard specifies the method of liquid chromatography in food natamycin.
This standard applies to the determination of food natamycin.
The standard quantitative detection limit of 0.5mg/kg (solid and semi-solid samples), 0.5mg/L (liquid sample).
Principle 2
Samples were extracted with methanol, the use of water freezing or centrifugation to remove the fat content in the sample purification way, reversed-phase liquid chromatography - ultraviolet detection

3 Reagents and materials
3.1 Water. deionized water or equivalent purity.
3.2 methanol. AR, chromatography.
3.3 Natamycin (natamycin). purity ≥95%.
3.4 standard stock solution. Accurately weigh an appropriate amount of natamycin standard (accurate to 0.1mg), dissolved in methanol, dubbed the concentration of
100μg/mL standard stock solution.
3.5 glacial acetic acid. pure class distinctions.
3.6 0.45μm, 0.22μm syringe filter (aqueous).
4 Instrument
4.1 High performance liquid chromatograph. with UV detector.
4.2 ultrasonic cleaner.
4.3 Electronic balance. a sense of the amount of 0.1mg.
4.4 syringe. one, 10mL.
4.5 funnel. a diameter of about 7cm.
4.6 Refrigerator. -20 ℃ ~ -15 ℃.
4.7 centrifuge.
5 Sample Preparation
5.1 Solid and semi-solid samples
5.1.1 cheese, ham, yogurt sample
10.00g accurately weighed sample was added 30mL of methanol ultrasonic extraction 30min, add water 10mL shake in the refrigerator freezer
1h, frozen samples were taken immediately after the filtration, the filtrate was set to room temperature, followed by over 0.45μm and 0.22μm syringe filter to collect

5.1.2 moon cake, pastry samples
Accurately weighed 5.00g moon cake skin, placed in a centrifuge tube, add 30mL of methanol ultrasonic extraction 30min, add water 10mL shake Set
In centrifuge at 3500r/min centrifugal 5min, the supernatant successively through 0.45μm and 0.22μm syringe filter, collecting the filtrate about
