This standard replaces GB/T 15400-1994 "Determination of tryptophan in feed by spectrophotometry".

1 Scope
This standard specifies high performance liquid chromatography and spectrophotometry for the determination of tryptophan in feed.
This standard applies to the determination of tryptophan in feed ingredients, compound feed, concentrated feed and concentrate supplements.
The limit of quantitation of high performance liquid chromatography is 0.02%; when the maximum sample weight of the sample is 700mg, and the dilution factor of the hydrolyzate is 25 times,
The limit of quantitation of the photometric method is 0.04%.

3.1 Principle
The tryptophan in the sample is hydrolyzed by lithium hydroxide solution, separated by high performance liquid chromatography, detected by ultraviolet or fluorescence detector, external standard method

3.2 Reagents or materials
Unless otherwise specified, the reagents used in this standard are of analytical grade.
3.2.1 Water. Meet the requirements of the first grade water in GB/T 6682.
3.2.2 Methanol. chromatographically pure.
