Fentanyl and five other fentanyl compounds in forensic water samples
Substance and metabolite analysis by liquid chromatography-mass spectrometry
1 Scope
This document specifies the presence of fentanyl, desphenethylfentanyl, acetylfentanyl, desphenethylfentanyl and fentanyl in water samples (sewage, surface water, etc.) in the field of forensic science.
Liquid chromatography-mass spectrometry (LC-MS) method for the determination of 4-anilino-N-phenethylpiperidine.
This document applies to the five fentanyl-like substances and their metabolites (hereinafter referred to as the “five fentanyl-like substances”) in water samples (sewage, surface water, etc.) in the field of forensic science.
Other samples can be used for reference.

4 Principle
Using blank samples and spiked samples as controls, the five target substances (basic information of compounds) in water samples were analyzed according to the requirements of parallel operation.
See Appendix A) for extraction, purification and concentration, and detection by liquid chromatography-mass spectrometry, based on retention time, mass spectrometry characteristic ion pairs and ion pair abundance.
The concentration ratio was used as the basis for qualitative judgment; the internal standard calibration curve method was used for quantitative analysis based on the chromatographic peak area ratio.
5 Reagents and Materials
5.1 Reagents
The water used in the experiment should comply with the first-grade water specified in GB/T 6682.Unless otherwise specified, all reagents used in the analysis were chromatographically pure.
Reagents include.
a) Concentrated hydrochloric acid. analytical grade, mass fraction 36%~38%;
b) Drinking water. tap water;
c) 5% ammonia methanol solution (taking 1000 mL as an example). measure 50 mL of concentrated ammonia (mass fraction 25%) and dilute it with methanol.
And make up to 1000mL, mix well;
d) 30% ammonium acetate solution (taking 100 mL as an example). weigh 30 g of ammonium acetate, dissolve it in water and make up to 100 mL, mix well;
e) 0.1% formic acid solution (taking 1000 mL as an example). Pipette 1.0 mL of formic acid, dilute with water and make up to 1000 mL, ultrasonically
