3 Principle

Agglutination method. A, B and H antigen substances exist on the surface of human red blood cells, under the action of anti-A antibody, anti-B antibody or anti-H antibody, An antigen-antibody reaction occurs to cause agglutination of red blood cells. The ABO blood type can be judged based on the agglutination reaction with the corresponding antibody. Dissociation method (protein sticking method). This method is an antigen-antibody complex formed by reacting an antigen and an antibody at a suitable temperature. Under the condition that the antigen-antibody complex can release the antibody while maintaining the original properties of the antigen and the antibody unchanged. In the sample In the presence of anti-A antibody or anti-B antibody, A and B blood-type substances undergo antigen-antibody reaction to form an antigen-antibody complex. When in At 50 ° C ~ 56 ° C, the antigen-antibody complex dissociates, leaving the antibody free in the liquid, when the temperature is reduced to about 37 ° C, free The antibody reacts with the corresponding indicator erythrocyte antigen, and agglutination can be observed under the microscope. According to the corresponding instructions In the case of agglutination reaction, the antigen carried by the blood mark can be inferred to determine the ABO blood type.
