1 Scope

This standard specifies the method for the determination of the total amount of tocopherol in the cosmetics and the content of α-tocopheryl acetate. This standard applies to the test of the total amount of tocopherol and α-tocopheryl acetate in cosmetics.

2 Principle

Extraction of α-tocopherol, β-tocopherol, γ-tocopherol, δ-tocopherol and α-fertility in cosmetics with a mixed solvent of ethanol, diethyl ether and n-hexane Phenol acetate, centrifugation, taking supernatant, using di-n-octyl phthalate as internal standard, determined by high performance liquid chromatography with UV detector, according to Its retention time is qualitative and the internal standard method is quantitative.

The reagents were of analytical grade unless otherwise stated. 3.1 Methanol, excellent grade, filtered with 0.45μm filter. 3.2 n-hexane. 3.3 Ethanol. 3.4 ether. 3.5 α-tocopherol, purity 96.9%. 3.6 β-tocopherol, purity 96.9%. 3.7 γ-tocopherol with a purity of 99.5%. 3.8 δ-tocopherol with a purity of 92.4%. 3.9 α-tocopherol acetate, purity 99.2%. 3.10 Di-n-octyl phthalate, purity 99.0%. 3.11 Tocopherol and α-tocopheryl acetate standard stock solution. weigh the appropriate amount (accurate to 0.1mg) α-tocopherol, β-tocopherol, γ-sheng Tocopherol, δ-tocopherol, α-tocopherol acetate, di-n-octyl phthalate, make up to 100mL with n-hexane, shake well, and prepare various substances A mixed standard stock solution with a concentration of 0.5 mg/mL was stored at 4 °C. 3.12 Di-n-octyl phthalate internal standard stock solution. weigh the appropriate amount of di-n-octyl phthalate (accurate to 0.1mg) in 100mL capacity In a measuring flask, add n-hexane to the mark, shake well, prepare a solution with a concentration of 3.3 mg/mL, and store at 4 °C. 3.13 Di-n-octyl phthalate internal standard solution. Take 5 mL of internal standard stock solution (3.12) in a 100 mL volumetric flask, dilute to n-hexane. Shake well (this solution has a shelf life of 1d).
