1 Scope
This standard specifies the Niucheng disease immune agar diffusion test method.
This standard applies to the quarantine of cattle and Ibaraki disease in and out of the country.
2 materials
2.1 Reagents
2.1.1 Agarose (for electrophoresis), 0.9% saline.
2.1.2 antigen, standard positive serum.
2.2 Equipment
2.2.1 Straws, graduated cylinders, flasks, 90 mm diameter plates.
2.2.2 Water platform, balance, metal puncher, adjustable micropipette and tip.
2.2.3 Template paper. Six holes around one hole in the middle, the hole diameter is 4mm; the hole spacing (edge) is 3mm.
2.3 Tested serum

3 method of operation
3.1 Preparation of agar plates
Take 1 g of agarose, 100 mL of 0.9% physiological saline, add to the Erlenmeyer flask, and fully boil and melt in a water bath, or 115 ° C for 10 min.
