Test guidelines on environmental safety assessment for chemical pesticides – Part 11. Silkworm acute toxicity test 1 Scope GB/T 31270 of this Part specifies the basic requirements such as material, condition, operation, quality control, data processing, test report of silkworm acute toxicity test, measured by leaf dipping method and fumigation method. This Part applies to silkworm leaf dipping and fumigation toxic test, providing a reference for other types of pesticide. This Part does not apply to volatile and insoluble chemical pesticides. 2 Terms and definitions For the purpose of this Part, the following terms and definitions shall apply. 2.1 Medium lethal concentration In leaf dipping method test, test substance concentration that causes 50% death of test organism, indicated as LC50. Note. unit as mg a.i./L. 2.2 Test substance Substance that needs to be tested in test. 2.3 Chemical pesticide Pesticide is an artificial pesticide by using chemical substances, in which biomimetic synthesis pesticide is made by taking active substance of natural products as matrix, conducting the imitation, structure-change and innovation. Synonymy. Synthetic organic pesticide [NY/T 1667.1 -2008, definition 2.3.1] 2.4 Technical material In the process of manufacturing, the final product that is obtained by combination of effective elements and impurities. It shall not include visual foreign matter and any additives; if necessary, a small number of stabilizer shall be added. [NY/T 1667.2-2008, definition 2.5.1] 2.5 Formulation products Products with stable service condition that are made from pesticide, original medicine and additive. [NY/T 1667.2-2008, definition 2.4] 2.6 Active ingredient. a. i. Active ingredient refers to special chemical structural ingredient with biological activity. [NY/T 1667.2-2008, definition 3.1] 2.7 Reference substances Chemical substance or mixture that is used to prove or deny certain property of test substance or judge effectiveness of test system. 3 Test summary 3.1 Method summary Silkworm acute toxicity test includes leaf dipping toxic method and fumigation method. Choose relevant methods to implement test according to pesticide registration management rules and other provisions. 3.2 Leaf dipping toxic test Adopt the solution with different concentrations to dip folium mori; after drying, it shall be used to feed silkworm. During the whole process of feeding and treatment mori, toxic symptom and death status of silkworm in test shall be observed after 24 h, 48 h, 72 h and 96 h. After test finishes, analyze the data and calculate medium lethal concentration LC50. 3.3 Fumigation toxic test Fumigation toxic test is a test, based on health, using medicine to imitate indoor drug delivery condition; it shall be implemented in fumigation room or fumigation test device in accordance with test requirements. On the basis of meeting test requirements, relevant Annex A (Informative) Division of toxic level of pesticide to silkworm Acute toxicity of pesticide can be divided into four levels according to toxic medium lethal concentration LC50 (96h), by acute leaf dipping method, as shown in Table A.1 Table A.1 Division of toxic level of pesticide to silkworm Level of toxicity LC50 (96 h)/(mg a. i. /L) Severe toxic LC50 ≤ 0.5 Highly toxic 0.5 < LC50 ≤ 20 Medium Toxic 20 < LC50 ≤ 200 low toxic LC50 > 200 Fumigation toxic test is a test, based on health, using medicine to imitate indoor drug delivery condition. If death rate of silkworm is above 10%, then it is deemed that it is high risk to silkworm. ......
