1 Scope
This standard specifies the herbal medicines and products of quintozene residues in gas chromatography - mass spectrometry method.
This standard applies to ginseng, ginseng oral liquid quintozene residues determination.
2 Method summary
Sample with n-hexane - acetone solution was extracted, concentrated sulfuric acid sulfonation, by gas chromatography - mass spectrometry, external standard.
3 Reagents and materials
Unless otherwise specified, reagents were of analytical grade water as a water.
3.1 hexane. chromatographically pure.
3.2 acetone. chromatography.
3.3 Concentrated sulfuric acid. pure class distinctions.
3.4 anhydrous sodium. Burning 4h at 650 ℃, placed in a desiccator.
3.5 sulfate solution. 40g/L.
3.6 PCNB (Quintozene, C6Cl5NO2, CAS. 82-68-8) Standard. 99% or greater purity.
3.7 PCNB standard solution. Accurately weigh an adequate amount of PCNB standards, formulated with n-hexane at a concentration of 100μg/mL standard
Quasi stock solution. Needed with n-hexane diluted to the appropriate concentration of standard working solution. Stored at 0 ℃ ~ 4 ℃ refrigerator.
3.8 anhydrous sodium sulfate column. 80mm × 40mm (inner diameter) cylindrical funnel, bottom cushion height about 5mm cotton, refill 10g anhydrous sodium sulfate
(3.4).
4 instruments and equipment
4.1 Gas chromatography - mass spectrometry with electron impact source (EI).
4.2 vortex mixer.
4.3 high-speed homogenizer. 24000r/min.
4.4 Centrifuge. 3000r/min.
4.5 rotary evaporator.
4.6 mill herbs.
5 Sample Preparation and Storage
Take a representative sample of ginseng about 500g, fully crushed through 2.0mm sieve. Take a representative sample of about oral ginseng 500g, mix
Uniform, loaded into a clean container and sealed and labeled, stored refrigerated at 4 ℃.
In the course of sampling and sample preparation operation, should prevent sample contamination or change PCNB residue content occur.
