1 Scope

This standard specifies the method for determination of sorbic acid in meat and meat products. This standard applies to the testing of sorbic acid in meat and meat products.

2 Principle

The sorbic acid in the sample was extracted, purified with neutral alumina, and then subjected to high performance liquid chromatography.

3 reagents and materials

The reagents used below are analytically pure reagents unless otherwise specified, and the water is re-distilled water. 3.1 Methanol. excellent grade pure. 3.2 Ammonium acetate. 3.3 Neutral alumina. for chromatography, (100 ~.200) mesh. 3.4 0.02mol/L ammonium acetate solution. 1.54g ammonium acetate was weighed, dissolved in water and diluted to 1L. 3.5 Sorbic acid. 99% (National Center for Standard Materials Research). 3.6 Standard stock solution of sorbic acid. Accurately weigh 25.0mg sorbic acid standard, dissolve it with a small amount of ethanol, dilute with water and dilute to volume 25.0 mL, shake well. This solution contained 1.0 mg of sorbic acid per ml. Stored in the refrigerator for 15 days. 3.7 Standard solution of sorbic acid. Pipette 5.0 mL of standard stock solution of sorbic acid in a 100 mL volumetric flask, dilute to the mark with water, and Shake well. This solution contained 50 μg of sorbic acid per ml. Stored in the refrigerator, valid for 7 days. 3.8 Sorbic acid standard series solution. 1.0, 2.0, 3.0, 4.0, 5.0mL sorbic acid standard intermediate solution in 10mL volumetric flask Add water to the scale and shake well. That is, a standard series solution containing 5.0, 10.0, 15.0, 20.0, 25.0 μg of sorbic acid per ml is obtained.

4 Instruments and equipment

5 Determination steps

5.1 Sample processing Take.200g sample and grind it, weigh about 1.00g (accurate to 0.01g) sample, put it in 50mL plastic test tube, add 5mL water, spin The mixture was vortexed for 1 min, ultrasonically extracted for 15 min, and then centrifuged at 4000 r/min for 20 min. Column chromatography. use a 10mL syringe, fill a small amount of absorbent cotton at the bottom, and contain 3cm high neutral alumina, first wet the neutral oxygen with water.
